模拟失重大鼠血液流变学异常与肌肉退行性变化的中药防护

【摘要】 目的观察四组中药合剂对尾吊大鼠血液流变学异常和肌肉退行性变的防护作用.方法采用大鼠头低位30°尾部悬吊模型模拟失重对机体的影响.120只SD雄性大鼠随机分为6组:对照组、单纯尾吊组、丹黄刺组(尾吊同时给予丹参、黄芪和刺五加生药的水煎剂)、参川熟组(尾吊同时给予川芎生药、熟地和人参地上甙的水煎剂)、参山杜组(尾吊同时给予西洋参、山楂、杜仲生药的水煎剂)、参银生组(尾吊同时给予人参根、银杏叶、生地的水煎剂). 实验第30d,经心脏取血,检测全血粘度、红细胞变形性、纤维蛋白原含量、血细胞压积、红细胞形态、肌肉重量、肌纤维类型和面积等指标.结果单纯尾吊导致大鼠体重明显降低(p<0.001),肌肉重量、肌肉纤维面积、Ⅰ型纤维比例显著下降(p<0.05,0.01),中切和低切血粘度、纤维蛋白原含量、异形红细胞比例明显增高(p<0.001);红细胞最大变形指数(DImax)和积分变形指数(IDI)(p<0.001,0.01)、血细胞压积明显降低(p<0.001).中药合剂组与单纯尾吊组相比, 血液流变学指标有较大改善,并可显著增加尾吊大鼠腓肠肌重量(p<0.05).结论中药合剂可改善尾吊导致的血液流变学及肌肉组织改变.

【Abstract】 Objective To investigate the role of Chinese herb medicine to counter the abnormity of blood circulation and the muscle atrophy caused by simulated weightlessness. Methods 120 rats were divided into 6 groups: control group, head down tills - 30° group (HDT), DanHuangci group (HDT + Dan Sheng, Huang Qi, Ci Wu Jia), ShenChuanShu group (HOT + ginseng, Chuan Xiong, Shu Di), ShenShanDu group (HDT + American ginseng, Haw, Du Zhong ), ShenYingSheng group (HDT+ ginseng, the leaves of gingkgo, Sheng Di). The body weight, erythrocyte deformability (DImax, IDI), erythrocyle shape, hemalocrit(HCT), blood viscosity, plasma Cbrinogen, blood cell aggregation index, muscle weight, the type and size of muscle fiber were observed following HDT for 30 days. Results After 30 days - HDT, the body weight of rats, muscle weight,size of muscle fiber and the percentage of I type muscle fiber decreased distinctly (p <0.05, 0.01, 0.001) . The blood viscosity in low and middle shear, plasma fibrinogen and the rate of erythrocytes with abnormal shape increased significantly due to HDT ( p < 0.001) , and erythrocyte deformability (DImax, IDI) and hematocrit(HCT) decreased distincdy (p < 0.001 ,p < 0.01) . Chinese herb medicine were available to improve the hemorheological parameters, and increased the weight of gastrocnemius under simulated weightlessness (p < 0.05). Conclusions It is suggested that Chinese herb medicine are effective to meliorate the changes of blood circulation and muscle under simulated weightlessness.
