低细胞密度状态下的人骨髓间质干细胞成骨能力研究

【摘要】 目的 研究低密度人骨髓间质干细胞(Humanbonemarrow -derivedmesenchymalstemcells,hMSCs)体内成骨能力及影响因素。方法 从人髂骨骨髓分离培养hMSCs。以不同细胞密度与块状和颗粒型磷酸钙陶瓷(HA/TCP)载体混合,移植于裸小鼠体内,分别观察2和3个月,组织学评价研究hMSCs异体体内成骨的最小细胞密度。按3.3×1 0 6个细胞/cm3 载体的比例混合细胞与载体(HA/TCP和鼠尾胶原包被的HA/TCP) ,体外共培养一周后移植裸鼠皮下,3个月后,组织学评价细胞载体体系体外培养和胶原包被载体对hMSCs体内成骨能力的影响。结果 只有以3.3×1 0 6个细胞/cm3 载体在体内移植3个月后可观察到在块状和颗粒HA/TCP表面有骨组织生成。用鼠尾胶原包被的颗粒HA/TCP和细胞与载体体外共培养并无明显促进骨组织生成作用。结论 保证hMSCs异体成骨的最小细胞密度为3.3×1 0 6个细胞/cm3 载体。在低密度hMSCs状态下,鼠尾胶原和细胞与载体体外共培养并不能提高hMSCs的成骨能力。

【Abstract】 Objective To study the osteogenic capability of low-density human bone marrow-derived mesenchymal stem cells (hMSCs).Methods hMSCs were separated from human iliac crest marrow and cultured. Different density hMSCs were mixed with block and particle HA/TCP vehicles. Then the mixtures were transplanted subcutaneous part of nude mice. Two and three months later, histological evaluation of all in vivo transplantation grafts were performed in order to determine how many hMSCs per cubic centimeter and how long the grafts could form new bone tissue. Particle HA/TCP was coated by collagen deriving from rat tail. hMSCs were mixed with the coated vehicles and cultured in osteoinductive medium one week. Then the grafts were transplanted subcutaneously in nude mice. Three months later, histological evaluation of theexperiment’s results determined the effect of collagen and co-culture of hMSCs and vehicles one week on hMSCs’ osteogenesis.Results hMSCs were cultured and identified successfully. Histological evaluation’s results showed that bone formation was found only in 3 months grafts of 3.3×10 6 hMSCs per cm3 in HA/TCP group. The new bone adhered to the surface of HA/TCP and appeared lamellar-like structure. Under low-density hMSCs, The HA/TCP coated with collagen and co-culture of hMSCs and vehicles didn’t obviously promote hMSCs’ osteogenesis by statistical test.Conclusion To guarantee to form bone tissue by hMSCs, the minimum hMSCs density should be 3.3×10 6 hMSCs per cm 3 HA/TCP. Under low-density hMSCs, HA/TCP coated with collagen and co-culture of hMSCs and vehicles can’t obviously increase hMSCs’ osteogenesis.
