牵引成骨过程骨生长因子表达的定量分析
The quantitative analysis of cytokines during distraction osteogenesis
【摘要】 目的探讨牵引成骨过程骨生长因子的变化与牵引成骨机制的关系。方法采用ELISA、放射免疫等方法定量分析骨痂内各种细胞生长因子的含量,采用免疫组织化学方法观察生长因子在骨组织内的分布。结果牵引成骨过程BMP - 2、TGF - β1、IGF -Ⅱ以及BGP的表达均增强;牵引成骨区、骨吸收区均有TGF - β1分布;术后早期BMP - 2在截骨区分布相对较少,随着术后时间的延长,BMP - 2的分布趋于密集,特别是成骨细胞聚集的部位;而随着骨的成熟,TGF - β1、BMP - 2的分布减少。结论牵引成骨过程有较多骨组织形成的机制可能与骨生长因子的高表达有关。
【Abstract】 Objective To investigate the quantitative change in bone growth factor during distraction osteogenesis and to study the relationship between the mechanism of distraction osteogenesis and the changes in bone growth factor.Methods The quantitative change in bone growth factor was analyzed by ELISA and radioimmunoassay methods. The distribution of bone growth factor in bone was observed with histoimmunochemical methods.Results The content of BMP-2?TGF-β1?IGF-Ⅱ and BGP in the callus increased during distraction osteogenesis. TGF-β1 was distributed at the area of osteogenesis and bone absorption. BMP-2 was distributed at the area of osteogenesis, especially at the site of osteoblastic congregation.Conclusion The mechanism of distraction osteogenesis may relate to a high expression of bone growth factor.
