应用辣根过氧化物酶逆行示踪技术观察左右侧脊神经节间纤维联系
Observation of connections between the fibers of the left and right spinal ganglion neurons with retrograde tracing technique of horseradish peroxidase
【摘要】 目的:观察大鼠左右侧脊神经节之间的纤维联系,以寻找两侧脊神经节 (背根节)的神经元交互支配这一电生理学现象的形态学基础。方法:实验于2004-11/2005-03在首都医科大学人体解剖学系解剖学实验室进行,取Wistar大鼠48只,根据注射示踪剂的不同随机分为3 组:游离辣根过氧化物酶(300-500g/L)组,麦芽凝集素结合辣根过氧化物酶(10-50 g/L)组和霍乱毒素B单位结合辣根过氧化物酶(3g/L) 组,按注射部位不同又分为左侧脊髓(L3或L4)、左侧脊神经节(L3或L4)、左侧坐骨神经和左侧股神经4小组,每小组4只,注射量均为3μL。注射示踪剂后两三天麻醉状态下处死取材,钨酸钠法显色。观察各组动物左右侧脊神经节、脊髓组织切片阳性标记的细胞及纤维。结果:48只全部进入结果分析。无论注射哪种标记物,一侧神经、神经节注射辣根过氧化物酶仅在单侧前角有标记细胞,背外侧束、后角、中央管后灰质处有标记的纤维,个别对侧可见标记的纤维,偶见一两个标记细胞。脊髓一侧注射的除同侧有标记细胞外,对侧(脊髓、神经节)也有少量标记细胞。结论:一侧背根节细胞的中枢突可达到两侧后角,不能直接定论是否存在或不存在直接联系,还需要进行深入的电镜和神经递质研究。
【Abstract】 AIM:To study the connections between the left and right spinal ganglion neurons in rats,in order to search the basic morphological rules for the electrophysiological phenomenon:reciprocal innervation of the bilateral spinal ganglion (dorsal root ganglion) neurons. METHODS:The experiment was conducted at laboratory of Department of Human Anatomy of Capital University of Medical Sciences from November 2004 to March 2005.Forty-eight Wistar rats were gained and were randomly divided into 3 groups according to different injection tracer: free horseradish peroxidase group (300-500 g/L),wheat germ agglutinin-horseradish peroxidase conjugate group (10-50 g/L)and cholera toxin B-horseradish peroxidase conjugate group (3 g/L). Then they were also assigned into 4 groups: the spinal cord ( L3 or L4), spinal ganglion (L3 or L4), sciatic nerve and femoral nerve in the left side based on different injection part with 4 rats in each group,with the volume of injection of 3 μL.After being injected with tracer for 2-3 days the animals were killed under drugged stale.and were reacted with sodium wolfamate staining method. Cells and fibers with positive tag of spinal ganglia and tissue section of spinal cord in the left and right sides of animals in every group were observed. RESULTS:Forty-eight rats were all involved in the result analysis.The marked cells were found in unilateral anterior horn after injecting horseradish peroxidase into nerve and ganglions no matter injection with which kind of marker. There were marked fibers at dorsolateral tract, posterior horn, post-gray of central canal and special fibers in contralateral part; a few marked cells were found accidentally. Injecting into one side of spinal cord, there were tagged cells in the same side, also a few in contralateral part (spinal cord and ganglions). CONCLUSION:The central process of cells at one-side dorsal root ganglion can reach to the posterior horn of both sides,but it cannot be concluded whether exists the direct connections, and deep researches with electron microscope and neurotransmitter are needed.
