丹皮酚对佐剂性关节炎大鼠的炎症反应及细胞因子水平的干预作用
Inflammatory reaction of paeonal in rats with adjuvant arthritis and intervention of cytokines
【摘要】 目的:探讨丹皮酚对佐剂性关节炎大鼠的抗炎作用及其细胞因子与炎症的关系,为临床治疗风湿及类风湿性关节炎提供理论依据。方法:实验于2003-10-01/12-15在潍坊医学院药理实验室完成。取健康成年Wistar大鼠50只,雌雄各半,随机均分为对照组,模型组,模型+地塞米松组,模型+丹皮酚1组,模型+丹皮酚2组。后4组每只大鼠分别于右足垫皮内注射弗氏完全佐剂0.05mL造模型;造模后第17天开始,模型+地塞米松组大鼠腹腔内注射地塞米松2.5mg/(kg·d),模型+丹皮酚1组和模型+丹皮酚2组分别灌胃丹皮酚100,200mg/(kg·d),连用10d。观察丹皮酚对佐剂性关节炎大鼠炎症的作用,并用放射免疫法测定其血清中白细胞介素1,2,6,及肿瘤坏死因子的含量。结果:实验大鼠50只均进入结果分析。①与对照组比较,模型组大鼠足爪肿胀度及全身病变都非常明显;与模型组比较,模型+地塞米松组,模型+丹皮酚1组,模型+丹皮酚2组大鼠的足爪肿胀度及全身病变均明显减轻;但丹皮酚小剂量作用弱。②模型组与对照组比较,白细胞介素1,6及肿瘤坏死因子的含量增加[对照组:(0.26±0.02)mg/L,(83.6±4.70),(0.81±0.06)mg/L;模型组:(0.35±0.02)mg/L,(140.15±13.9),(0.92±0.09)mg/L,P<0.01或0.05],白细胞介素2含量无明显变化(P>0.05);模型+地塞米松组、模型+丹皮酚1组、模型+丹皮酚2组与模型组比较,白细胞介素1,2,6,肿瘤坏死因子含量都显著降低[模型+地塞米松组:(0.21±0.03)mg/L,(3.70±0.51),(89.06±7.24),(0.57±0.06)mg/L;模型+丹皮酚1组:(0.25±0.03)mg/L,(4.25±0.37),(107.52±6.25),(0.71±0.13)mg/L;模型+丹皮酚2组:(0.24±0.02)mg/L,(3.81±45),(96.32±4.41),(0.64±0.16)mg/L,P<0.01];但模型+丹皮酚1组较模型+丹皮酚2组作用弱,模型+丹皮酚1组与模型+丹皮酚2组作用均较模型+地塞米松组弱。结论:丹皮酚明显减轻佐剂性关节炎大鼠的炎症反应,降低其血清中白细胞介素1,2,6及肿瘤坏死因子的含量,有剂量依赖性,作用较地塞米松弱。丹皮酚的抗炎作用与减少炎症因子白细胞介素1,6有关,也与降低免疫促进因子白细胞介素2、肿瘤坏死因子的含量而抑制免疫反应有关。
【Abstract】 AIM: To observe the anti-inflammatory effects of paeonal in adjuvant arthritis rats and relation between cytokine and inflammation, and provide theoretical evidence for rheumatism and atrophic arthritis in clinical therapy. METHODS: The experiment was conducted at the Department of Pharmacology, Weifang Medical College from October to December 2003. Fifty healthy adult female and male (half and half) Wistar rats were selected, and assigned randomly into control group, model group, model + desamethasone group, first model + paeonal group and second model + paeonal. The rats in the latter 4 groups were treated with 0.05 mL Freund’s complete adjuvant with intradermal injection at right foot pad, respectively to built models. From the 17th day after building models, the rats in the model + desamethasone group were treated with 2.5 mg/kg desamethasone per day through intraperitoneal injection. Those in the first model + paeonal group and second model + paeonal group were given 100 mg/kg and 200 mg/kg paeonal per day with gastric perfusion for 10 days. The anti-inflammatory effects of paeonal in adjuvant arthritis rats were observed. The contents of interleukin 1, 2 and 6 with tumor necrosis factor (TNF) were detected with radio-immunity method. RESULTS: Fifty experimental rats were all involved in the result analysis. 1 Compared with the control group, the foot swelling and lesion at total body were all extremely significance in rats of model group; Compared with model group, the foot swelling and lesion at total body in rats relieved obviously in the model + desamethasone group, first model + paeonal and second model + paeonal group. But the effects of small dosage of paeonal were weak. 2 Comparison between model group and control group, contents of interleukin 1 and 6 with TNF increased control group: (0.26 ±0.02) mg/L,(83.6±4.70),(0.81±0.06) mg/L;model group:(0.35 ±0.02) mg/L,(140.15±13.9),(0.92±0.09) mg/L,P < 0.01 or 0.05 . There was insignificant change of content of interleukin 2 (P > 0.05); Comparison of model + desamethasone group, first model + paeonal group and second model + paeonal group with model group, the contents of interleukin 1, 2 and 6 with TNF all reduced markedly model + desamethasone group:(0.21±0.03) mg/L,(3.70±0.51),(89.06±7.24),(0.57±0.06)mg/L; first model +paeonal group:(0.25±0.03)mg/L,(4.25± 0.37),(107.52±6.25),(0.71±0.13)mg/L; second model + paeonal group:(0.24±0.02)mg/L,(3.81±45),(96.32±4.41),(0.64±0.16) mg/L,P < 0.01 . But the effects on first model + paeonal group were weak as compared with the second model + paeonal group. The effects on first model + paeonal group and second model + paeonal group were weak as compared with the model + desamethasone group. CONCLUSION: The paeonal lightens significantly the inflammatory reaction in adjuvant arthritis rats, decrease the contents of interleukin 1, 2 and 6 with TNF in serum with dose-dependent effects, and its effect is weak as compared with desamethasone. The anti-inflammatory effects of paeonal are related with the decrease of, anti-inflammatory factors, interleukin 1 and 6, and also related with decrease of, immunological enhancement factor, interleukin 2, and content of TNF to inhibit the immunologic reaction.
