穴位敷贴对支气管哮喘豚鼠肿瘤坏死因子及白细胞介素2的影响 空白对照与标准对照
Effect of acupoint application on tumor necrosis factor and interleukin-2 of guinea pig with bronchial asthma blank and standard control
【摘要】 目的:观察中药穴位敷贴对实验性哮喘豚鼠血清中哮喘相关细胞因子肿瘤坏死因子及白细胞介素2的影响,并与地塞米松的作用效应相比较。方法:实验于2004-05/06在广州中医药大学动物实验中心完成。选用健康Hartly豚鼠60只,随机分为4组,每组各15只。采用卵蛋白致敏制作豚鼠支气管哮喘模型,用中药穴位敷贴治疗(穴位敷贴组)与地塞米松组(标准对照)及模型组(空白对照)和正常对照组比较。①穴位敷贴组于造模成功,哮喘发作次日开始治疗。治疗穴位选择大椎、肺俞、肾俞。用大块胶布将药物敷贴固定于穴位处。每次敷药6h,隔日治疗次,共治疗7次。②地塞米松组采用腹腔注射地塞米松0.5mg/kg,治疗时间与次数同敷贴组。③模型组造模成功后令其自然恢复,正常对照组不造模,不给予任何治疗。④各组动物治疗结束后血清肿瘤坏死因子和白细胞介素2水平的检测采用放射免疫分析法。结果:在造模及治疗阶段模型组死亡4只、穴位敷贴组死亡3只,地塞米松组死亡2只,正常对照组死亡2只。每组取10只共40只进入结果分析。①模型组豚鼠血清肿瘤坏死因子、白细胞介素2水平均高于正常对照组犤(0.707±0.113),(0.377±0.134)mg/L;(1.552±0.315)(0.790±0.311)mg/L,P<0.05犦。②穴位敷贴组和地塞米松组血清肿瘤坏死因子、白细胞介素2水平明显低于模型组犤(0.445±0.195),(0.33±0.132),(0.707±0.113)mg/L;(1.033±0.541),(0.971±0.502),(1.55±0.315)mg/L,P分别小于0.05和0.01犦。③穴位敷贴组和地塞米松组肿瘤坏死因子、白细胞介素2水平差异不显著犤(0.445±0.195),(0.33±0.132)mg/L;(1.033±0.541),(0.971±0.502)mg/L,P>0.05犦。结论:穴位敷贴与地塞米松均可降低血清肿瘤坏死因子、白细胞介素水平,但差异不显著,说明穴位敷贴与地塞米松的标准作用一致。穴位敷贴治疗哮喘可能是通过降低血清肿瘤坏死因子、白细胞介素2水平这些炎症因子而发挥起效应。
【Abstract】 AIM To observe the influence of acupoint application on the asthma related cytokines of tumor necrosis factor and interleukin 2 in serum of experimental guinea pigs with asthma and compare its role and effect with those of dexamethasone. METHODS The experiment was carried out in the animal experimental center of Guangzhou University of Traditional Chinese Medicine between May and June 2004. Sixty healthy Hartly guinea pigs were randomly divided into four groups with 15 guinea pigs in each group. Guinea pigs were made into models of bronchial asthma by means of albumin sensitization The levels of tumor necrosis factor and interleukin 2 in those treated with Chinese herb acupoint application acupoint application group dexamethasone group standard control group and model group blank control group were compared with those in the normal control group. ① After the successful establishment of models guinea pigs in the acupoint application group were treated from the next day of asthma attack The herbs were fixed at the acupoints of Tachui Feishu and Shenshu with rubberized cloth of large pieces 6 hours for each time once every other day for totally 7 times. ② Guinea pigs in the dexamethasone group were treated with intraperitoneal injection of dexamethasone 0.5 mg/kg the therapeutic time and times were the same as those in the @ cupoint application group. ③ Guinea pigs in the model group recovered naturally after the successful establishment of models Those in the normal control group were not made into models and received not any treatment. ④ After treatment the serum levels of tumor necrosis factor and interleukin 2 were detected with radioimmunoassay in each group. RESULTS During model establishment and treatment 4 guinea pigs in the model group 3 from the acupoint application group 2 in the dexamethasone group and 2 in the normal control group died respectively Finally 10 guinea pigs were selected in each group and involved in the analysis of results. ① The serum levels of tumor necrosis factor and interleukin 2 were higher in the model group than in the normal control group 0.707±0.113 0.377 ±0.134 mg/L 1.552±0.315 0.790±0.311 mg/L P < 0.05. ② The serum levels of tumor necrosis factor and interleukin 2 were obviously lower in the acupoint application group and dexamethasone group than in the model group 0.445±0.195 0.330±0.132 0.707±0.113 mg/L 1.033±0.541 0.971±0.502 1.552±0.315 mg/L P < 0.05 and 0.01. ③ The serum levels of tumor necrosis factor and interleukin 2 were insignificantly different between the acupoint application group and dexamethasone group 0.445±0.195 0.330±0.132 mg/L 1.033±0.541 0.971±0.502 mg/L P > 0.05. CONCLUSION Both acupoint application and dexamethasone can decrease the levels of tumor necrosis factor and interleukin 2 in serum without significant differencesindicating that the standard effects of acupoint application and dexamethasone are concordant.Acupoint application may play its role in treating asthma by decreasing the serum levels of the inflammatory factors of tumor necrosis factor and interleukin 2.
