年龄因素对蛋白脂蛋白139~151诱导自身免疫性脑脊髓炎小鼠模型的影响
Effect of age on SJL/J mice model with inductive autoimmune encephalomyelitis due to proteinlipid protein 139-151
【摘要】 目的:观察蛋白脂蛋白139~151(PLP139~151)诱导的不同周龄实验性自身免疫性脑脊髓炎小鼠在发病时间及神经功能评分方面的差异。方法:实验于2004-06/11在苏州大学附属一院神经病学实验室进行。选用SJL/J雌性小鼠40只,分为6周龄组、8周龄组、10周龄组及12周龄组,每组10只。分别对各组小鼠接种蛋白脂蛋白139~151,观察其首次发病情况,并进行神经功能评分。于免疫后45d取脑和脊髓做组织病理学检查,分别行苏木精-伊红和固兰染色,观察炎症浸润及髓鞘脱失情况。结果:进入结果分析为37只小鼠。①6,8,10及12周龄组小鼠分别于免疫后(7.9±0.9)d,(10.1±0.9)d,(13.4±1.0)d及(17.1±1.0)d发病,两两比较差异显著(Sig.=0.000,P<0.05),低年龄组小鼠发病明显早于高年龄组。②在病情的严重程度上,6周龄组明显重于12周龄组[(4.5±1.1)分,(3.0±1.4)分,(Sig.=0.025,P<0.05)]。至免疫后45d时6周龄组神经功能评分明显低于12周龄组[(1.7±0.8)分,(2.6±0.9)分,(Sig.=0.022,P<0.05)]。各组之间在炎症浸润及髓鞘脱失程度上差异无明显区别(P>0.05)。结论:不同周龄的SJL/J小鼠在蛋白脂蛋白139~151免疫后发病时间各不相同,且首次发病的严重程度也有差异。小鼠年龄是影响实验性自身免疫性脑脊髓炎模型建立的关键因素。
【Abstract】 AIM:To observe whether there are some differences in the day of disease onset and the evaluation of neural function in the first attack of experimental autoimmune encephalomyelitis(EAE) induced by proteinlipid protein 139-151(PLP139-151)in SJL/J mice at different ages.METHODS:The experiment was carried out in laboratory of neurology, the First Affiliated Hospital of Soochow University.Forty SJL/J female mice were selected and were divided into sixweekold group,eightweekold group, tenweekold group and twelveweekold group with ten mice in each group. PLP139-151 was incubated in SJL/J mice to observe the first onset and score the neural function.Fortyfive days after immunization,brain tissue and spinal cord tissue were obtained to perform histological and pathological examination.Inflammatory infiltration and demyelination were observed by HE staining. RESULTS:Thirtyseven mice in sixweekold group,eightweekold group, tenweekold group and twelveweekold group fell ill at(7.9±0.9)days,(10.1±0.9)days,(13.4±1.0)days and(17.1±1.0)days, respectively , after immunity. There was significant difference in the day of disease onset among the four groups(Sig.=0.000,P< 0.05).The day of EAE onset in low age groups was significantly earlier than that in old age groups.The scores on the severity of disease in 6weekold mice were higher than those in 12weekold mice[(4.5±1.1),(3.0±1.4)points,(Sig.=0.025, P< 0.05)].The scores on neural function in 6weekold mice were lower than those in 12weekold mice[(1.7±0.8),(2.6±0.9)points,(Sig.=0.022,P< 0.05)].There was no obvious difference in the inflammatory infiltration and demyelination among the four groups at 45 days after immunization.CONCLUTION:The day of onset and the severity of EAE induced by PLP139-151 are different in SJL/J mice at different ages.The age of the animal plays an important role in EAE induction.
