地黄多糖诱导骨髓间充质干细胞分化为神经样细胞的效应

【摘要】 目的:探讨地黄多糖对骨髓间充质干细胞分化为神经细胞的诱导效应。方法:实验于2004-03/06在湖南省中医学院内科实验室进行,取1月龄SD大鼠1只,取股骨和胫骨,利用贴壁法分离纯化骨髓间充质干细胞。将骨髓间充质干细胞分为3组:地黄多糖组用地黄多糖诱导其向神经细胞分化,β-巯基乙醇组β-巯基乙醇诱导,对照组不做任何处理。光镜下观察细胞形态,采用免疫细胞化学方法鉴定神经细胞特异性抗原标志神经丝蛋白和神经胶质细胞特异性抗原胶质纤维酸性蛋白的表达。结果:地黄多糖组经地黄多糖诱导培养24h后细胞显示为典型的神经细胞样形态,神经丝蛋白阳性为(82.3±6.1)%,胶质纤维酸性蛋白阳性细胞为(10.5±2.1)%。β-巯基乙醇组的细胞神经丝蛋白及胶质纤维酸性蛋白阳性细胞数较少,分别为(22.3±4.7)%,(42.3±5.3)%。对照组神经丝蛋白及胶质纤维酸性蛋白染色均为阴性。结论:地黄多糖具有诱导骨髓间充质干细胞分化为神经样细胞的作用,其机制需探讨。

【Abstract】 AIM:To study the effect of rchmannia glutnosa polysaccharides on differentiation of rat bone marrow mesenchymal stem cells into nerve cells.METHODS: The experiment was finished in the Laboratory of Department of Internal Medicine, Hunan College of Traditional Chinese Medicine from March to June 2004.Femur and tibia from a one month rat were selected.Pure bone marrow mesenchymal stem cells were separated with differential adheion and divided into three groups:rchmannia glutnosa polysaccharides group was induced by rchmannia glutnosa polysaccharides to differentiate into nerve cells;β mercaptoethanol group was induced by β mercaptoethanol, and control group received no administration. The morphological changes of the cells were observed under phase contrast microscope.The neuronal specific markers neurofilement protein and glial fibriliary acidic protein were evaluated by using indirect immunocytochemisty staining.RESULTS: After induction of 24 hours, the differentiated cells showed typical neuronal morphology, and expressed neuron specific markers:neurofilement protein [(82.3± 6.1)% ] and glial fibriliary acidic protein[(10.5± 2.1)% ] in rchmannia glutnosa polysaccharides group, which were lower in β mercaptoethanol group [(22.3± 4.7)% , (42.3± 5.3)% ]. There was no positive expression of neurofilement protein and glial fibriliary acidic protein in control group. CONCLUSION: Rchmannia glutnosa polysaccharides can induce bone marrow mesenchymal stem cells to differentiate into nerve cells, but the mechanism need further research.
