电针激活老年痴呆大鼠海马蛋白激酶信号通路的作用
Effect on the signal pathway of hippocampal protein kinase in senile dementia ra ts with the activation of electroacupuncture
【摘要】 目的:探讨电针能否激活老年痴呆大鼠海马蛋白激酶信号传导通路,从而提高学习记忆能力。方法:实验于2003-04-25/05-30在成都中医药大学针灸推拿学院时间生物实验室进行。取SD大鼠100只,先以迷宫测试筛选60只,随机分为正常组、模型组、假手术组、电针组,每组15只;以穹隆-海马伞损伤进行老年痴呆造模,造模成功后,每组随机选取9只接受相应的处理:正常组、模型组和假手术组接受与电针组相同方式和相同时间的抓取、固定;电针组穴位选取“百会”、“涌泉”、“太溪”、“血海”,电针频率20Hz,1次/d,30min/次;20次后,放免法分别测定每组大鼠海马组织细胞胞膜蛋白激酶C(proteinC,PKC)、酪氨酸蛋白激酶(proteintyrosinekinase,PTK)含量。结果:正常组、模型组、假手术组、电针组PKC活性分别为5.13±0.58),((4.92±0.93),(3.98±0.75),(5.05±0.83)nkat/g,其他3组与模型组比较,差异有非常显著性意义(F=4.182,P<0.01);4组PTK活性分别为(0.477±0.063),(0.476±0.014),(0.369±0.080),(0.468±0.110)nkat/g,模型组与其他组比较,差异有显著性意义P(<0.05)。结论:穹隆-海马伞损伤所致老年痴呆大鼠可能存在海马蛋白激酶信号通路的抑制,电针能够激活低下的蛋白激酶信号通路,从而提高老年痴呆大鼠习记忆能力。
【Abstract】 AIM:To investigate whether electroacupuncture therapy can active the signal pa thway of hippocampal protein kinase in senile dementia rats,so as to improve the ability of learning and memory. METHODS:The experiment was performed in the Laboratory of Time and Biology,Col lege of Acupuncture and Massage,Chengdu University of Traditional Chinese Medici ne from April 25 to May 30 in 2003. Among 100 SD rats,60 rats were selected with maze test and randomly divided into normal group, model group,sham operation g roup and electroacupuncture group with 15 rats in each group.The models of senil e dementia were established with the injury of fornix fimbria of hippocampus.Af ter that,9 rats were randomly selected from every group and treated as follow:Ra ts in normal, model and sham operation groups were snatched and fixed as the sa me types and the same time as those in electroacupuncture group.In electroacupun cture group,the acupoints were Paihui(Du20),Yungch’uan(Jing Well Point,K1),T’ai hsi (Shu Stream and Yuan Source Point,K3)and Xuehai(Sp1),and the frequency was 20 Hz which was lasting 30 minutes once a day.After twenty treatments,the conte nt of protein kinase C(PKC) and tyrosine protein kinase(PTK) of cell membrane in rats hippocampal tissue were assayed with radio immunoassay. RESULTS:PKC activity was(5.13±0.58) nkat/g in normal group,(4.92±0.93) nkat/ g in model group,(3.98±0.75) nkat/g in sham operation group and(5.05±0.83) nk at/g in electroacupuncture group. That of model group was significant difference compared with other three groups(F=4.182, P< 0.01).PTK activity was(0.477±0.06 3) nkat/g in normal group,(0.476±0.014) nkat/g in model group,(0.369±0.080) nk at/g in sham operation group and(0.468±0.110) nkat/g in electroacupuncture gro up.That of model group was significant difference compared with other three grou ps (P< 0.05). CONCLUSION:The signal pathway of hippocampal protein kinase can be inhibited i n senile dementia rats with the injury of fornix fimbria of hippocampus,but the electroacupuncture therapy can active the signal pathway of hippocampal protein kinase,so as to improve the ability of learning and memory of senile dementia r ats.
