临产前后母血、羊水EGF、PGE2变化及其对胎盘组织bcl-2表达的影响
The effects of the concentrations of EGF and PGE2 in maternal blood and amniotic fluid on the expression of bcl-2 in placenta on labor onset.
【摘要】 目的:探讨临产前后母血、羊水中表皮生长因子(EGF)、前列腺素E(PGE2 )浓度的变化及其对胎盘滋养层细胞bcl 2蛋白表达的影响。方法:足月妊娠53例分成两组,临产组28例,未临产组25例。用放射免疫法测定孕妇母血、羊水中EGF、PGE2 的浓度,用免疫组化法测定胎盘滋养层细胞bcl- 2蛋白的表达。结果: (1)临产组母血、羊水中EGF、PGE2 水平均高于未临产组(P<0 .05); (2)临产组胎盘滋养层细胞bcl -2的蛋白表达明显低于未临产组(P<0. 05); (3)母血、羊水中EGF的浓度与PGE2 的浓度变化呈明显的正相关。母血、羊水中EGF、PGE2 的浓度变化与bcl- 2蛋白表达呈负相关。结论:EGF及bcl- 2介导的胎盘滋养层细胞凋亡在启动分娩发动过程中具有重要作用;且EGF可能通过PGE2影响bcl- 2的表达。
【Abstract】 Objective:To investigate the changes of the concentrations of epidermal growth factor (EGF) and prostaglandins E 2 (PGE 2) in maternal blood and amniotic fluid on laber onset,and their influences on the expression of bcl-2 proteins in placental trophoblastic cells.Methods:Fifty-three women in term pregnancy were divided into two groups.One group on the time of labor onset were twenty-eight women.Another group not on the time of labor onset were twenty-five women.The concentrations of EGF and PGE 2 in maternal blood and amniotic fluid were detected by radioimmune assay.The expression of bcl-2 protein in placental trophoblastic cells was detected by immunohistochemical assay.Results: (1) The concentrations of EGF and PGE 2 in maternal blood and amniotic fluid in labor onset group were higher than those of not labor onset group(P<0.05).(2)The expression of bcl-2 protein in labor onset group was significantly lower than that of the group not labor onset( P<0.05).(3) There was a positive correlation between the concentrations of EGF and PGE 2 in maternal blood and amniotic fluid, but the expression of bcl-2 protein had a negative correlation with the concentrations of EGF and PGE 2 respectively.Conclusion: It is considered that EGF and the apoptosis of placental trophoblastic cells induced by bcl-2 may play an important role in the initiation of labor onset.EGF may influence the expression of bcl-2 by PGE 2.
