纸色谱分离-紫外分光光度法测定当归中阿魏酸含量

【摘要】 直接用紫外分光光度法测定当归提取液中阿魏酸含量时,由于杂质干扰较大,影响测定精度,而采用纸层析分离可以将杂质除去。研究对纸层析的展开系统、不同萃取溶剂的稳定性等因素进行比较分析后,建立了纸层析分离-紫外分光光度法测定当归中阿魏酸含量的方法。纸层析分离条件为:以氯仿、甲醇、水和冰醋酸按体积比2∶2∶6∶0.5配比作为展开剂,检测波长为330nm,加样回收率为99.61%,标准差为2.79%,吸光度在0.020~0.061时线性关系良好。

【Abstract】 Determining precision was affected by the impurities in the extract of Angelica sinensis when the content of ferulic acid was determined directly by UV spectrophotometry.Paper chromatography (PC) was used to eliminate these impurities.The separation condition of PC and the stability of different extraction solvents were studied.Based on the results,a determination method of ferulic acid in Angelica sinensis by PC spectrophotometry was established.The elution system for PC was:chloroform:methanol (water:acetic acid (6∶2∶2∶0.5),the wavelength for spectrophotometry was 330 nm.The recovery rate of the method was 99.61% and relative standard deviation ( RSD ) was 2.79%.The absorbency shows good linearity relation between 0.020 and 0.061.
