GDNF和HSV-GDNF对体外培养大鼠脊髓运动神经元损伤后Bcl-2表达的影响
Effects of GDNF and HSV-GDNF on Bcl-2 expression of spinal cord motoneurons of rats after their scratch injury in vitro
【摘要】 目的:观察GDNF和单纯疱疹病毒载体介导的GDNF(HSV-GDNF)对体外培养大鼠脊髓运动神经元损伤后Bcl-2表达的影响。方法:将培养8d的神经元,随机分成4组,行划痕损伤后观察神经元存活数、Bcl-2免疫反应(BCL-2-IR)阳性神经元数目和平均光密度。结果:损伤后第1、3、5和7天,GDNF和HSV-GDNF组的三项指标均明显高于同期对照组;第1、3天,GDNF功效优于HSV-GDNF;第5、7天,HSV-GDNF的功效较好。结论:GDNF和HSV-GDNF具有增强受损运动神经元Bcl-2表达的作用,且HSV-GDNF作用时间较长。
【Abstract】 Objective:To study the effects of GDNF and HSV-GDNF on Bcl-2 expression of spinal cord motoneurons of rats after being injured in vitro. Methods: The neurons cultured in vitro were randomly divided into four groups on the 8th day. The number of survival neurons, and the number and the mean optical densities of Bcl-2 immunoreactive positive neurons were respectively observed after scratch injury. Results:By drawing a comparison with corresponding samples, it showed that these items in GDNF and HSV-GDNF group were significantly improved on the 1st, 3rd, 5th and 7th day. The effects of GDNF were better than those of HSV-GDNF on the 1st and 3rd day, however, they were just converse on the 5th and 7th day. Conclusion: GDNF and HSV-GDNF can enhance Bcl-2 expression and HSV-GDNF has a longer effect than GDNF on injured motoneurons.
