重组大肠杆菌产β-胡萝卜素的发酵条件

【摘要】 对重组大肠杆菌产-胡萝卜素发酵条件进行了研究.在M9培养基中添加Span-200.7g/L有利于细胞生长和β-胡萝卜素表达,初始pH值6.0、接种体积分数5%、发酵温度28℃、抗生素氨苄青霉素质量浓度80μg/mL和氯霉素质量浓度40μg/mL为最佳发酵条件,在7L发酵罐进行发酵动力学试验表明,重组菌最适发酵周期为22h,细胞干重最高可达1.55μg/L,β-胡萝卜素表达量可达0.75μg/mL.

【Abstract】 The fermentation conditions of β-carotene production by recombinant Escherichia coli were studied. Addition of 0. 7 g/L into M9 medium were favorable to cell growth and β-carotene expression. The optimum fermentation conditions were as follows: initial pH 6. 0, inoculation volume 5%, temperature 28℃, Amp concentration 80 μg/mL and Cm concentration 40μg/mL. Under these conditions, the fermentation kinetics test was determined in 7 L fermentor. The results showed that the maximum cell dry weight was 1. 55 g/L, the β-carotene concentration was 0. 75μg/mL, and the optimum fermentation time was 22 hours in the fermentor.
