【摘要】 目的研究抑癌基因Phosphataseandtensinhomologydeletedonchromosometen(PTEN)在口腔鳞癌组织中的表达情况及其临床病理学意义,探讨PTEN与口腔鳞癌发生发展的关系及作用机制,为利用基因治疗口腔鳞癌提供理论和实验依据。方法应用免疫组织化学S-P法检测34例口腔鳞癌组织中PTEN蛋白的表达,另有10例正常口腔黏膜组织作为对照组。结果正常口腔黏膜组织中PTEN蛋白全部表达,阳性率为100%;口腔鳞癌组织中PTEN蛋白的阳性表达率67.6%,显著低于正常口腔黏膜组织;PTEN在高、中、低分化口腔鳞癌组的阳性表达率分别为83.3%、78.6%和25%,差异有统计学意义(P<0.05);PTEN在口腔鳞癌组织中的蛋白表达与患者的年龄、性别无显著相关性(P>0.05)。结论PTEN基因在口腔鳞癌的发生发展过程起一定作用;PTEN蛋白的表达与患者的年龄、性别无明显相关性,而与组织分化程度显著相关。更多还原

【Abstract】 Objective To study the protein expression of PTEN gene in oral squamous cell carcinoma(OSCC) and its clinicopathological significance,and to investigate the role of PTEN in the process of OSCC and the relationship between PTEN and development of OSCC,providing the theoretical and experimental basis for gene treatment of oral cancer.Methods The expression of tumor suppressor gene PTEN was examined in 34 OSCC specimens using SP immunohistochemical technique.Control study was made in 10 subjects with normal oral mucosa(NOM).Results The expression level of PTEN in OSCC (67.6%) was lower than that in NOM(100%).The positive rates of PTEN were 83.3% and 78.6% in highly and mederately differentiated OSCC but 25% in poorly differentiated OSCC(P<0.05).The expression of PTEN protein was not related to age or sex(P>0.05).Conclusions PTEN gene plays a role in the development of OSCC.The level of PTEN protein is related to the degree of differentiation in OSCC.更多还原
