P27、P53及FAK的表达与肝癌肝移植后肿瘤复发与转移关系的研究

【摘要】 目的 探讨P2 7、P5 3及粘着斑激酶 (FAK)的表达与原发性肝细胞性肝癌 (HCC)行肝移植治疗后肿瘤复发与转移的关系。方法 应用免疫组织化学方法 ,检测其切除的肝癌标本中P2 7、P5 3与FAK的表达 ,分析其表达与移植后肿瘤复发与转移之间的关系和意义。结果 P2 7与P5 3的表达同中晚期肿瘤复发与转移具有一定的关系 ,有明显的协同作用 (趋性 χ2 检验 ,P<0 . 0 5 ) ;而FAK的表达在两组间无差异 (P <0 . 0 5 )。结论 P2 7低表达的HCC行肝移植后肿瘤更易于复发与转移 ,P2 7可能是评价预后的重要指标 ,P2 7与P5 3的联合检测更有助于预测移植后肿瘤的复发与转移。

【Abstract】 Objective To study the expressions of protein P27,P53 and focal adhesion kinase (FAK) in hepatocellular carcinoma (HCC)and their significance in the recurrence and metastasis of HCC for patients who received liver transplantation.Methods Thirty patients with HCC who underwent liver transplantation in this hospital were divided into two groups according to the time of recurrence and metastasis after transplantation: recurrence and metastasis in early stage(≤6 months,RMES) and in later stage(>6 months,RMMLS). Immunohistochemistry was performed to investigate the expressions of P27, P53 and FAK in HCC samples from the patients,and the relationship between the expressions of P27, P53, FAK and the recurrence and metastasis of HCC was analysed.Results Expression levels of P27 and P53 correlated with RMMLS. The expression of P27 was significantly lower in the group of RMES (P<0.05), and no significant difference was found for the expression of P53 between RMES and RMMLS(P>0.05);combined analysis of P27 and P53 showed synergetic effect for RMMLS(P<0.05 for trend analysis).No significant difference in expression of FAK was found between the two groups(P>0.05).Conclusions Lowered expression of P27 may contribute to early recurrence and metastasis of HCC after liver transplantation.P27 may be an important marker for evaluation of the prognosis of HCC patients after liver transplantation.Combined analysis of P27 and P53 is helpful for evaluation of recurrence and metastasis of HCC after liver transplantation.
