骨形成肽羧基端衍生物G35I对成骨细胞和破骨细胞的体外影响
The Effects of Osteogenic Growth Peptide C-terminal Derivative G35I on the Osteoblast and Osteoclast in Vitro
【摘要】 目的:探讨骨形成肽羧基端衍生物G35I在体外对成骨细胞和破骨细胞的影响。方法:利用体外人成骨细胞和骨髓来源大鼠破骨细胞培养和RT-PCR方法进行研究。结果:(1)G35I在体外可以刺激人胚胎长骨来源的成骨细胞的增生。(2)G35I在10-7mol/L可以刺激骨髓来源的破骨细胞样细胞的诱导分化形成,而在其他浓度(10-8~10-11mol/L)此作用不明显。(3)G35I在10-7mol/L条件下可以抑制成骨细胞骨保护素的表达,且在72h作用最明显。结论:骨形成肽羧基端衍生物G35I在不同浓度时对骨代谢的两种功能细胞均有一定作用。
【Abstract】 Objective Bone remodeling was carried out by osteoclast and osteoblast. The effects of osteogenic growth peptide C-terminal derivative G35I on osteoblast and osteoclast were investigated. Methods The effects of G35I on the forming of osteoclastic like cell induced from the bone marrow cells and on the human osteoblast were investigated in vitro. Semi-quantity RT-PCR was used to examine the effect of G35I on the expression of osteoprotegerin in human osteoblast. Results 1 G35I may stimulate the proliferation of human osteoblast in vitro. 2 G35I may enhance the formation of osteoclastic like cell at the concentration of 10-7 mol/L in bone marrow cells culture system. 3 G35I may decrease the expression of osteoprogerin in the human osteoblast in culture at the concentration of 10-7 mol/L and the effect was more significant when the action went on for 72 h. Conclusion Osteogenic growth peptide C-terminal derivative G35I may have effects on both osteoblast and osteoclast at different concertration.
