核因子κB和uPA在人膀胱移行细胞癌中的表达及临床意义

【摘要】 目的 探讨膀胱移行细胞癌组织中核因子κB (NF -κB) 和尿激酶纤溶酶原激活剂(uPA) 的表达及相互关系。方法 采用免疫组化二步法对38例膀胱移行细胞癌和10 例非癌膀胱粘膜标本中NF -κB p65 和uPA蛋白的表达进行测定, 分析其与肿瘤临床病理学特征的关系及内在联系。结果 NF- κB p65和uPA蛋白在膀胱移行细胞癌中的表达显著高于非癌膀胱粘膜(均P<0. 01); NF -κB p65 和uPA蛋白在膀胱移行细胞癌中的表达具有相关关系(r=0 .89, P<0 .01); NF κB蛋白的组成性活化与膀胱癌淋巴结转移(P<0 .01) 和病理分级(P<0 .05) 有关; uPA蛋白的表达与膀胱癌浸润深度(P<0. 05)、病理分级(P<0 .05) 和淋巴结转移(P<0 .01) 有关。结论 NF- κB的组成性活化和uPA的高表达与膀胱癌的临床病理学特征有关, NF -κB可能通过调控uPA的合成促进肿瘤的进展, 可能是未来膀胱癌治疗中的一个重要靶点。

【Abstract】 Objective To investigate the expression of NF-κB p65 and uPA protein and their relationships in transitional cell carcinoma (TCCs) of bladder. Methods The expression of NF-κB and uPA protein in 38 cases of TCCs of bladder and 10 cases of non-carcinoma bladder mucosa were detected by immunohistochemical technique and its relation with tumor clinicopathological characteristics was analyzed. Results The expression levels of NF-κB and uPA protein in TCCs of bladder were higher than those in non-carcinoma bladder mucosa (both P<0.01). There was significant correlation between the expression of NF-κB and uPA in TCCs of bladder. NF-κB activation was correlated with tumor invasion-related clinicopathological features such as lymph node metastasis (P<0.01),high pathological grade (P<0.05). The overexpression of uPA protein was correlated with clinicopathological characteristics of bladder cancer such as lymph node metastasis (P<0.01),clinical stage (P<0.05),pathological grade (P<0.05). Conclusion Constitutive activation of NF-κB and the overexpression of uPA were correlated with biological behaviors of TCCs of bladder and suggested that NF-κB activity was related to tumor progression due to its transcriptional regulation of uPA and NF-κB may be an important target for the development of future anti-invasion therapy in cases of TCCs of bladder.
