肝炎平对家兔血吸虫肝纤维化MMP-2、TIMP-1表达的影响

【摘要】 目的 研究肝炎平抗家兔血吸虫肝纤维化的作用, 探讨其抗肝纤维化的可能机制。方法 实验分为3 组:正常对照组(N组)、血吸虫感染+吡喹酮组(A组) 及血吸虫感染+吡喹酮+肝炎平治疗组(B组)。采用苏木精伊红染色及Masson染色, 观察肝组织病理学改变; 采用免疫组织化学SP 法检测各组肝组织基质金属蛋白酶2(MMP-2)、金属蛋白酶组织抑制因子- 1 (TIMP- 1) 的表达水平。结果 B组肝组织胶原纤维含量低于A组(P<0 05); B组MMP 2、TIMP -1的表达水平均低于A组(P<0 .01, P<0 .05)。结论 在抗血吸虫治疗后使用肝炎平,能通过减少肝组织MMP- 2、TIMP- 1的表达而发挥抗血吸虫肝纤维化的作用。

【Abstract】 Objective To study the antifibrotic effect and possible mechanism of Ganyanping on schistosomal liver fibrosis of rabbits after anthelmintic treatment.Methods The rabbits were randomly divided into 3 groups: normal control group (N), schistosomal infection+praziquantel group (A) and schistosomal infection+praziquantel+Ganyanping group (B). The histopathological changes of liver were observed by HE and Masson staining. The expression of matrix metalloproteinase-2 (MMP-2) and tissue inhibitor of metalloproteinase-1 (TIMP-1) in liver tissue was detected by immunochemistry.Results The contents of collagenous fibers in group B were lower than in group A (P<0.05). The expression of MMP-2 and TIMP-1 was decreased in group B as compared to group A (P<0.01,P<0.05).Conclusion Ganyanping,after anthelmintic treatment,could reduce the expression of MMP-2 and TIMP-1 in liver of schistosomal liver fibrosis,which may be one of the antifibrotic mechanisms of Ganyanping.
