搭桥术中从胸骨和肋骨骨髓中富集CD34~+干细胞的方法
A Method to Enrich Human Bone Marrow CD34+ Stem Cells from Breast and Costal Bone During Coronary Artery Bypass Graft
【摘要】 目的为便于进行心肌干细胞移植的临床研究,探索一种搭桥术中从胸骨和肋骨骨髓中快速富集CD34+干细胞的方法。方法搭桥手术患者11例,男6例,女5例,平均年龄(65.4±8.9)岁,平均体重(60.8±14.6)kg。术中从胸骨和肋股骨髓中多点抽取骨髓,MiniMACS分选系统分离纯化骨髓CD34+干细胞,流式细胞术检测富集的CD34+细胞纯度,镜检细胞活力,计数富集干细胞的时间。结果本方法抽取的骨髓液20~30ml,可富集(1.7±0.8)×107个CD34+干细胞,纯度(95.8±1.6)%,活力(95.3±1.6)%;从抽取骨髓到富集到可用的CD34+细胞用时3.5~4h。结论搭桥术中从胸骨和肋骨骨髓中可快速富集理想的CD34+干细胞,获取细胞隐蔽,患者无痛苦,分选时间短,可在搭桥手术结束前提供心肌注射使用,为心肌干细胞移植的临床试验提供依据。
【Abstract】 Objective To explore a method to enrich human bone marrow CD 34+ stem cells from breast and costal bone during coronary artery bypass graft (CABG) in order for clinic studies of myocardium stem cell transplantation.Methods Eleven patients with coronary artery disease who should be done CABG, 6 male and 5 female with age mean (65.4±8.9) years and weight mean (60.8±14.6) kg, were studied. Bone marrow was taken from breast and costal bone during CABG. Cell purity was detected with flow cytometry, cell ability was examined with light microscope, and the time of enrichment was calculated.Results Twenty to thirty milliliter bone marrow was taken from each patient, the number of CD34+ stem cells was over (1.7±0.8) ×107, cell purity (95.8±1.6)%, cell ability (95.3±1.6)%, and the time of enrichment was about 3.5~4 h.Conclusion The ideal and good quality human bone marrow CD34+ stem cells from breast and costal bone can be enriched fast during CABG. Harvesting process was blanket and short, the patients had no complaints. These cells should be used before end of CABG. Experiment results were going to be used for myocardium cell transplantation clinic studies.
