酞酸二丁酯对黑腹果蝇寿命及SOD和MDA的影响
Effects of DBP on Life-span, Superoxide Dismutase Activity and Malondialdehyde Content in Drosophila Melanogaster
【摘要】 目的研究环境污染物酞酸二丁酯(Dinbutylphthalate,DBP)对黑腹果蝇寿命及抗氧化系统的影响。方法用生存试验检测果蝇寿命,记录不同剂量DBP染毒组果蝇生存数和死亡数,并测定DBP不同染毒浓度和不同染毒时间果蝇体内超氧化物歧化酶(superoxidedismutase,SOD)活性及丙二醛(malondialdehyde,MDA)含量。结果染毒后各剂量组雌雄果蝇半数死亡时间均缩短,平均寿命及平均最高寿命与对照组相比均有显著性差异(P<0.05或P<0.01),并呈现出明显的剂量-反应关系。不同浓度染毒组雌雄果蝇与对照组相比,SOD活性显著降低(P<0.05),MDA含量显著升高(P<0.05);雌雄果蝇在不同染毒时间的SOD活性和MDA含量与对照组相比差异均有显著性(P<0.01或P<0.05)。结论DBP可引起果蝇体内SOD活性下降,MDA含量上升,破坏果蝇体内的抗氧化系统,进而使果蝇寿命缩短,有很强的促衰老作用。
【Abstract】 Objective To study the effects of di-n-butyl phthalate (DBP) on life-span and system of anti-oxidation in drosophila melanogaster.Methods The drosophila’s life-span was detected with survival test, and the superoxide dismutase (SOD) activity and malondialdehyde (MDA) content in drosophila given different doses of DBP were detected at different time.Results DBP shortened drosophila average life-span and maximum average life-span significantly. Meanwhile, SOD activity dramatically decreased and MDA content remarkably increased with the increase of DBP.Conclusion DBP can significantly decreased SOD activity and increased MDA content, and then the system of anti- oxidation in drosophila was damaged, so that the drosophila’s life-span was shortened.
