高效液相色谱法测定复方磺胺甲噁唑片中磺胺甲噁唑和甲氧苄啶的含量

【摘要】 目的:建立同时测定复方磺胺甲唑片中磺胺甲唑和甲氧苄啶含量的高效液相色谱法。方法:采用KromasilC18色谱柱(150mm×4.6mm,5μm),甲醇-水-冰醋酸-三乙胺(225∶345∶0.5∶0.2)为流动相,流速为0.7ml/min,检测波长为239nm。结果:结果磺胺甲唑在15~75μg/ml范围内,峰面积与其浓度线性关系良好(r=0.9994),平均回收率为99.4%,重复性实验RSD为0.8%(n=6),检测限为0.1μg/ml;甲氧苄啶在3~15μg/ml的范围内,峰面积与其浓度线性关系良好(r=0.9992),平均回收率为99.1%,重复性试验RSD为0.6%(n=6),检测限为0.02μg/ml。结论:该法准确、简便、快速、专属性强,灵敏度高,适用于复方磺胺甲唑片的含量测定。

【Abstract】 Objective:To establish an HPLC method for determination of SMZ and TMP in Compound Sulfamethoxazole and Trimethoprim Tablets.Methods:SMZ and TMP in the tablets were determined by HPLC with a Kromasil C 18 column(250 mm×4.6 mm,5 μm) with UV detection at 239 nm, and the mobile phase was consisted of methyl alcohol-water-acetic acid glacial-triethylamine(225∶345∶0.5∶0.2), the flow rate was 0.7 ml/min.Results:SMZ and TMP in compound sulfamethoxazole and trimethoprim tablets can be separated. The calibration curve was linear (r=0.999 4) within the range of 15~75 μg/ml for SMZ, the average recovery was 99.4% with repeatability precision RSD was 0.8% (n=6). The calibration curve was linear (r=0.999 2) within the range of 3~15 μg/ml for TMP, the average recovery was 99.1% with repeatability precision RSD was 0.6% (n=6).Conclusion:The method is simple, quick and accurate with the high sensitivity and is suitable for the quality control for SMZ and TMP in compound sulfamethoxazole and trimethoprim tablets.
