抑制Smad7的表达对肾小管上皮细胞的影响

【摘要】 目的:本研究旨在观察血管紧张素Ⅱ(AngⅡ)诱导肾小管上皮细胞Smad7表达变化,以及对其凋亡及增殖的影响。方法:大鼠肾小管上皮细胞株NRK-52E分别与终浓度为0(对照组)、10-6mol/L、10-7mol/L、10-8mol/L、10-9mol/LAngⅡ共培养24h。通过AnnexinV-FITC/PIKit用流式细胞仪检测细胞凋亡指数,用免疫组化的方法检测增殖性细胞核抗原(PCNA)和Smad7蛋白的表达,RT-PCR检测Smad7mRNA的表达水平。结果:随着AngⅡ浓度的增加,10-6mol/LAngⅡ组与对照组相比凋亡指数显著增加,分别为(27.06±1.14)%vs(3.09±0.73)%(P<0.001)。与对照组相比,Smad7的表达在含有AngⅡ组中均呈现出低表达水平,并在一定范围内随AngⅡ浓度的增加而逐渐受到抑制,并与细胞凋亡指数成负相关(r=-0.82,P<0.05)。结论:AngⅡ可以抑制肾小管上皮细胞Smad7的表达,同时促进细胞凋亡,二者具有密切关系。

【Abstract】 Objective To investigate the Smad7 protein and mRNA levels in the etiology of Angiotensin Ⅱ (AngⅡ )- induced apoptosis in renal tubular epithelial cells. Methods Cells were incubated with the final concentration of AngⅡ was 0, 10- 6 mol/L, 10- 7 mol/L, 10- 8 mol/L and 10- 9 mol/L respectively. The apoptosis index was measured by using Annexin V- FITC/PI Kit with flow cytometer. Expression of Smad7 mRNA and protein were evaluated by semiquantitative reverse transcription- polymerase chain reaction(RT- PCR) and immunohistochemistry, respectively.Results The apoptosis index significantly increased in 10- 6 mol/L AngⅡ groups(27.06± 1.14)% vs (3.09± 0.73)% (P< 0.001). Ang Ⅱ stimulated the expression of Smad7 in renal tubular cells in a dose- dependent manner.Expression reduction of Smad7 was negatively correlated with Ang Ⅱ - induced renal tubular epithelial cells apoptosis(r=- 0.82,P< 0.01). Conclusion It is suggest that Smad7 play a very important role in AngⅡ - induced apoptosis in renal tubular epithelial cells.
