霉酚酸酯对2型糖尿病大鼠肾脏结缔组织生长因子表达的影响

【摘要】 目的:探讨霉酚酸酯对2型糖尿病大鼠肾脏结缔组织生长因子(CTGF)表达的影响。方法:将实验动物分为正常对照组、糖尿病组及霉酚酸酯治疗组。霉酚酸酯组予霉酚酸酯(MMF)15m g/(kg·d)治疗。13周后检测各组大鼠尿白蛋白排泄率,内生肌酐清除率,血糖,血脂,血胰岛素;H E及PA S染色观察肾脏病理改变;免疫组化方法检测肾组织中结缔组织生长因子(CTGF)、基质金属蛋白酶-9(MMP-9)及基质金属蛋白酶抑制剂-1(TIM P-1)的表达。结果:M M F可以减少2型糖尿病大鼠尿白蛋白排泄率及内生肌酐清除率;使大鼠肾脏组织中C TG F及TIM P-1的表达降低,M M P-9表达增加,并使肾脏病理改变好转。结论:霉酚酸酯对2型糖尿病大鼠肾脏有保护作用,其机制可能部分是通过下调2型糖尿病大鼠肾脏C TG F的表达及调节M M P-9和TIM P-1的平衡来实现的。

【Abstract】 Objective To investigate the effect of mycophenolate mofetil on renal connective tissue growth factor in type 2 diabetic rats.Methods Wistar rats were divided into three groups:normal control rats,diabetic rats,diabetic rats treated with mycophenolate mofetil (MMF)15 mg/(kg·d).Thirteen weeks later,we measured urinary albumin excretory rate,creatine clearance,blood glucose,blood insulin and blood lipid .Kidney pathology was observed. Inmmunohistochemistry was used to analyse the expression of CTGF,MMP 9 and TIMP 1. Results compared with the untreated diabetic group, urinary albumin excretory rate and creatinine clearance of MMF treated rats were substantially decreased .There were an decreasing expression of CTGF,TIMP 1 and increasing MMP 9 expression in the renal tissue of MMF treated group. MMF also improved the histological changes of diabetic rats. Conclusion mycophenolate mofetil has some renal protective effect on diabetic rats,partly through down regulating CTGF expression and regulating balance of MMP 9 and TIMP 1.
