【摘要】 目的探讨尿激酶激活剂抑制剂-1(PAI-1)对鼻咽癌侵袭和转移的影响。方法以高低不同淋巴道转移能力的鼻咽癌细胞系(CNE2Z)及其克隆株(CNE2ZH5,CNE2ZH59)作为研究材料;应用逆转录聚合酶反应(RTPCR)方法检测PAI-1表达水平;应用酶联吸附法(ELISA)测定PAI-1在3种细胞中的蛋白表达量;通过侵袭抑制实验观察PAI-1在鼻咽癌的侵袭和转移过程中的作用。结果PAI-1在CNE2Z、CNE2Z-H5中均有表达,但无显著性差异;PAI1在CNE2Z-H5-9中无表达。抗PAI-1抗体对CNE2Z-H5-9侵袭无影响。结论PAI1可能抑制鼻咽癌的侵袭和转移。更多还原

【Abstract】 Objective To explore the effect of PAI-1 on the invasion and metastasis in Nasopharyngeal Carcinoma(NPC).Methods A Nasopharyneal carcinoma cell-line(CNE-2Z) and its sub-line(CNE-2Z-H5,CNE-2Z-H5-9) with different ability of lymph node metastasis were used.RT-PCR was used to detect the expression of PAI-1 at mRNA level.The expression of PAI-1 at protein level was measured by ELISA.The effect of PAI-1 on the invasion and metastasis was observed in the invasion inhibitory experiments.Results The PAI-1 was expressed in CNE-2Z,CNE-2Z-H5 with no significant difference between them(P>0.05),but was not expressed in CNE-2Z-H5-9.Anti-PAI-1 antibody didn’t affect the invsion of CNE-2Z-H5-9.Conclusion PAI-1 may have an inhibitory effect on the metastasis and invasion in NPC.更多还原
