甲氟哌酸和氧氟哌酸的荧光胶束薄层测定研究

【摘要】 研究了甲氟哌酸(PFLX)和氧氟哌酸(OFLX)胶束薄层荧光法.以聚酰胺层析板为固定相,使用0.01mol/L的SDS和0.1mol/L的EDTA混合水溶液为展开剂,EDTA的加入避免了氟哌酸-金属离子的络合,使混合物得到较好的分离.在该条件下分离后的PFLX和OFLX的Rf值分别为0.60和0.77.试样斑点经薄层色谱扫描仪用荧光法扫描定量.激发波长为278nm(PFLX)和280nm(OFLX),截止波长在400nm.两个化合物的线性范围是0~90ng/斑点(PFLX)和0~110ng/斑点(OFLX),回归方程为C=3.21×10-3A-3.84(PFLX)和C=4.88×10-3A-3.54(OFLX),相关系数均在0.998以上.在血样和尿样中回收率为98.2%~108.1%,RSD<7.4%

【Abstract】 The simultaneous determination of Pefloxacin(PFLX) and Ofloxacin (OFLX) by thin layer chromatogrphy (TLC) was studied .A mobile phase of aqueous micelle solution of sodium dodecyl sulfate(SDS)∶EDTA=0.01∶0.1(mol/L∶mol/L),EDTA was used for the elimination of some transition metal ions to avoid the formation of norfloxacian-metal complexes.On polyamide thin layer sheet PFLX and OFLX were separated from each other,and their Rf values were 0.60 and 0.77,respectively.Fluorescence measurement was done using 278 nm (PFLX) and 280 nm(OFLX) as the excitation wavelength and the cut-off filter was set at 400 nm.The linear ranges were 0~90 ng(PFLX) and 0~110 ng(OFLX).The correlation coefficients of them were 0.998 and the linear regression equations were C=3.21×10(-3)A-(3.84) for PFLX and C=4.88×10(-3)A-3.54 for OFLX.In urine and serum samples the recoveries were (98.2 %)~108.1 %.
