肿瘤坏死因子-α和干扰素-γ对人嗜碱性粒细胞中Toll样受体基因表达的调节作用

【摘要】 目的:探讨肿瘤坏死因子α(TNFα)和干扰素γ(IFNγ)对人嗜碱性粒细胞中Toll样受体(TLR)基因表达的调节作用。方法:用逆转录聚合酶链反应检测人嗜碱性粒细胞系(KU812)中,TLR1~10mRNA的组成型表达;用实时定量聚合酶链反应检测TNFα或IFNγ刺激KU812细胞后,TLR1~10mRNA表达的变化。结果:人嗜碱性粒细胞有TLR1~10mRNA的组成型表达,其中TLR1~6和TLR9的mRNA表达量较高,而TLR7,8和TLR10mRNA的表达量相对较少。TNFα可明显上调TLR2~4mRNA的表达,而IFNγ对TLR2和TLR9的mRNA表达有明显的上调作用。结论:人嗜碱性粒细胞不仅参与了天然免疫,而且其免疫功能还受到细胞因子的调节。

【Abstract】 Objective:To investigate the regulatory effects of tumor necrosis factor-α(TNF-α)and interferon-γ(IFN-γ)on the expression of Toll like receptor(TLR)gene in human basophils.Methods:Constitutive expression of TLR1~10?mRNA in resting human basophil cell line(KU812)was examined with reverse transcriptase-polymerase chain reaction,while the alterations of TLR1~10?mRNA expression in KU812 cells were quantitated using real-time quantitative-polymerase chain reaction.Rusults:KU812 cells expressed all members of the TLR family(from 1 to 10),but mainly expressed TLR1~6 and TLR9.TNF-α strongly up-regulated the expression of TLR2~4?mRNA and IFN-γ significantly up-regulated mRNA expression of TLR2 and TLR9.Conclusion:The human basophils are involved in innate immune responses and their functions can be regulated by cytokines.
