添加液态烷烃氧载体对法夫酵母发酵生产虾青素的影响
Influences of Oxygen Vectors on Phaffia rhodozyma Growth and Carotenoid Production in Shake-flask Culture
【摘要】 在法夫酵母培养过程中,氧的供给是细胞生长和虾青素合成的重要因素。试验中测得法夫酵母摇瓶培养时比耗氧速率(qp)为60mg(O2)/(g.h)(cell),临界溶氧浓度10%。试验还考察了多种液态烷烃作为氧载体对法夫酵母生长和类胡萝卜素合成的影响,发现正十六烷作为氧载体效果较好,其最佳条件是在法夫酵母接种同时添加10%(v/v)的正十六烷。在此条件下,细胞干重达到10.1g/L,类胡萝卜素产量13.2mg/L,分别比对照增加22%和43%。同时,单位细胞类胡萝卜素产量从1.11mg/g增加到1.31mg/g,合成能力提高了18%。
【Abstract】 Oxygen supply plays a major role for Phaffia rhodozyma growth and carotenoid production. The specific oxygen uptake rate (qp) was measured as 60 mg (O2) /(g·h) (cell) and the critical dissolved oxygen was 10% of saturated oxygen during shake-flask culture. The influences of several hydrocarbons, as organic oxygen vectors, on P. rhodozyma growth and carotenoid production were investigated. It was found that hexadecane was the most suitable oxygen vector and optimum concentration was 10% (v/v). It this paper, the maximum biomass reached 10.1 g/L and carotenoid productivity reached 13.2 mg/L, which were increased 22% and 43% respectively compared to the control. It was also found that the specific carotenoid yield increased from 1.11 to 1.31 mg/g cell, which suggested that the synthesis capability was improved by 18%. These results showed that the use of oxygen vectors was an effective means to relieve oxygen deficiency in P. rhodozyma cultures and that carotenoid production.
