川金丝猴胰岛素样生长因子Ⅰ (IGF-Ⅰ)基因的克隆及序列分析
Cloning and Sequence Analysis of Golden Monkey (Rhinopithecus roxellana) Insulin-like Growth Factor Ⅰ Gene
【摘要】 本实验从金丝猴肝脏细胞RNA中反转录得到cDNA,利用设计的引物M1和M2扩增IGFⅠ基因,并将其克隆到pGEM-T载体上。经筛选、酶切、PCR鉴定、序列分析,证明该片段为金丝猴IGFⅠ基因的cDNA克隆。该片段由521个核苷酸组成,其中包括一个完整的开读框(ORF),编码一个由153个氨基酸组成的多肽。与GenBank中人、猪、鼠、马、牛、兔、山羊等哺乳动物的IGFⅠ序列比较,其编码成熟肽序列的同源性从93.8%(马)到88.6%(鼠),开放框序列的同源性从94.4%(人)到88.5%(鼠)。
【Abstract】 The insulin-like growth factor Ⅰ(IGF-Ⅰ)cDNA of the golden monkey (Rhinopithecus roxellana) was cloned for the first time from liver tissue by reverse transcription and amplified by PCR using a set of primers that we designed in this study. The amplified cDNA fragment was inserted into the vector pGEM-T. Through transferring, screening, enzymolysis and sequencing ,it was demonstrated that this fragment was IGF-Ⅰ gene cDNA. It is 521nts in length and contains one open reading frame (ORF), which encodes for a polypeptide containing 153 amino acids. The golden monkey IGF-Ⅰ gene has a high homology with other mammal IGF-Ⅰ genes.
