烫伤大鼠肠肌间神经丛Bax/Bcl-2的表达与大蒜素的干预

【摘要】 本研究观察了烫伤大鼠肠肌间神经丛Bax/Bcl-2蛋白的表达特征,并了解了大蒜素注射液的作用及作用机制。实验动物分成(1)对照组,(2)烫伤组,(3)药物组。(2)、(3)组又分烫伤后24、48、72h三个不同时间点。经麻醉、灌注、取材(空肠10cm)、后固定,制成铺片标本。采用免疫组织化学方法,对各时间点肠肌间神经丛进行Bax/Bcl-2检测。结果显示:(1)烫伤组Bax/Bcl-2的表达是以烫伤后24h阳性最强,48h有所下降,72h下降最明显;(2)药物组Bax阳性信号较对应时间点的烫伤组明显减弱,Bcl-2阳性信号较对应时间点的烫伤组明显增强。结果提示:(1)细胞凋亡是大鼠严重烫伤后小肠肌间神经丛神经元丢失的重要原因,而bax/bcl-2是参与神经细胞凋亡的重要凋控基因;(2)大蒜素注射液具有显著的保护作用,其机制可能与改善血循环,清除自由基,影响bax/bcl-2基因表达而起到抑制烫伤后迟发性神经元死亡有关。

【Abstract】 To observe the Bax/Bcl-2 protein expression in the intestinal myenteric plexus after the rat burn and protective mechanism of garlicin injection. Healthy SD rats were divided into control group, burned group and treated group. Each group was divided into three groups at 24, 48 or 72 hours after burn, the rats in each group were anaesthetized, prefused, and then the intestine was taken out. The intestine was fixed for 4 hours (4℃) again. Immunohistochemistry staining (sp method) was used to detect Bax/Bcl-2 protein expression in the intestinal myenteric plexus. The results showed that: (1) A significant increase in Bax/Bcl-2 expression was detected at 24 hours after burn, but it began to decrease at 48 hours after burn and was much lower at 72 hours after burn; (2) In the treated group(garlicin), the expression of the Bax was low and the expression of the Bcl-2 was stronger than that of burned group. The results suggest that: (1) The apoptosis is the main cause of the neuronal loss in the intestinal myenteric plexus after the severel burn, bax/bcl-2 is the significant regulating gene which participates in the apoptosis; (2) The garlicin has highly protective effects against apoptosis, and the protective mechanism might be related to improving the blood circulatory, eliminating oxygen free radicals and reducing the expression of Bcl-2 and Bax proteins.
