大林姬鼠中汉坦病毒的分离及M片断全基因序列分析
Sequence Analysis on Medium (M) Genome Segment of Hantavirus Isolated from Apodemus peninsulae in Northeast of China
【摘要】 目的 研究吉林省疫区大林姬鼠中携带的汉坦病毒的分子生物学特征,并探讨其分子进化规律。方法 应用直接免疫荧光、RT PCR法进行分型检测;阳性鼠肺标本用Vero- E6细胞分离病毒;测定汉坦病毒CJilin93株的M片断序列,并进行同源性比对和进化树分析。结果 从 58份大林姬鼠肺组织标本中共检测到 4个阳性标本,从中分离出 2株病毒:CJilin89和CJilin93;测序结果显示CJilin93株M基因全长 3 577bp,编码一个含有 1135氨基酸的G蛋白前体(GPC)。序列分析表明CJilin93株与分离自黑线姬鼠的Bao14株核苷酸序列同源性为 97 2%,氨基酸同源性为 99 4%;而与同样分离自俄罗斯远东地区大林姬鼠的AMR系病毒的同源性较远。系统发生树表明CJilin93株和Bao14株最为接近,共处于同一分支。结论 在俄罗斯远东地区和我国东北地区的大林姬鼠中携带不同种系的汉坦病毒。
【Abstract】 Objective To identify the molecular biological characteristics and to study the phylogenetic evolution of Hantavirus isolated from Apodemus peninsulae in Jilin,China.Method Lung samples were analysed by IFA and RT-PCR.Positive samples were isolated by Vero-E6 cell.Medium (M) genome segment of CJilin 93 strain was cloned,sequenced and analysed.Result The complete nucleotide sequence of the M genome of CJilin 93 strain included 3 577 bp in length encoded a GPC(glycoprotein precursor).Sequence comparison revealed close relation between CJilin 93 strain and Bao 14 strain carried by Apodemus agrarius.In contrast,the AMR strain harboured by Apodemus peninsulae showed a higher level of genetic diversity from CJilin 93 strain based on G2 segment alignment.Phylogenetic analysis showed that the CJilin 93 strain and Bao 14 strain formed a HTN clade.Conclusion Apodemus peninsulae carried HTN virus in the northeast of China.
