膀胱癌组织中上皮细胞钙依粘连蛋白基因启动子区CpG岛甲基化状态的分析
Analysis of CpG island methylation status of E-cadherin gene in bladder cancer
【摘要】 目的 探讨膀胱癌中肿瘤抑制基因上皮细胞钙依粘连蛋白(E cadherin)启动子区CpG岛的甲基化状 态及其与mRNA表达的关系。方法 收集30例新鲜的膀胱癌组织及其癌周正常组织;采用甲基化特异聚合酶 链反应(MSP)检测E cadherin基因启动子区CpG岛的甲基化状态;采用逆转录PCR检测E cadherin基因的mR NA表达。结果 E cadherin基因在膀胱癌组织中的甲基化阳性率(43.3%)显著高于在癌周正常组织中的甲基 化阳性率(6.7%,P<0.01);在复发性膀胱癌中的甲基化阳性率(64.7%)显著高于在原发性膀胱癌中的甲基化 阳性率(15.4%,P<0.05);在不同分级、分期膀胱癌中的差异无显著性。13例异常甲基化的膀胱癌组织中有10 例未检测到CDH1mRNA的表达,而13例未甲基化膀胱癌组织中有2例未检测到,二者差异有显著性(P< 0.01)。结论 E cadherin在膀胱癌组织中有一定程度的过甲基化,但与膀胱癌的临床分期和病理分级无显著相 关性,提示其可能参与了膀胱癌的发生;E cadherin基因CpG岛的过甲基化可能成为判断膀胱癌复发的指标之 一;E cadherin基因CpG岛的过甲基化可能是造成E cadherinmRNA不表达的原因之一。
【Abstract】 Objective To explore CpG island methylation status of tumor suppressor gene E-cadherin and its relation with mRNA in bladder cancer. Methods Thirty fresh bladder cancer tissues and paired adjacent normal tissues were obtained from cystectomy specimens. The CpG island methylation status of E-cadherin was examined by means of methylation specific PCR. The E-cadherin mRNA expression was investigated by means of reverse transcription PCR. Results The positive rate of E-cadherin methylation in tumor tissues was significantly higher than that in normal tissues (43.3% vs 6.7%,P<0.01). The positive rate in recurrent bladder cancer was significantly higher than that in primary bladder cancer(64.7% vs 15.4%,P<0.05). The methylation status did not correlate with stage and grade of bladder cancer. 10 of 13 methylated tumor tissues and 2 of 13 unmethylated tumor tissues did not show CDH1 mRNA expression(P<0.01) Conclusions There exists a certain degree of E-cadherin methylation in bladder cancer, but the methylation status does not correlate with grade and stage of bladder cancer, suggesting that the aberrant methylation of E-cadherin may be involved in the tumorigenesis of bladder cancer. The CpG island methylation status of E-cadherin may indicate the recurrence of bladder cancer, and may contribute to the loss of E-cadherin mRNA expression.
