糜酶对培养人血管平滑肌细胞血管紧张素Ⅱ生成及细胞增殖的作用

【摘要】 目的拟从细胞水平观察阻断糜酶途径后对人血管平滑肌细胞生成血管紧张素(AngⅡ)及细胞增殖的影响。方法通过植块培养法获得人脐动脉血管平滑肌细胞,并分组为:①DMEM组(空白培养液对照组);②AngⅠ组;③AngⅠ+卡托普利组;④AngⅠ+糜酶抑素组;⑤AngⅠ+缬沙坦组。分别阻断血素紧张素转换酶、糜酶和AngⅠ受体,进行细胞计数、细胞增殖指数及培养液中AngⅡ含量的测定。结果药物干预组较AngⅠ组细胞数量减少(P<0.01);糜酶抑素组与缬沙坦组能明显抑制细胞增殖(P<0.01);糜酶抑素组的AngⅡ含量明显降低(P<0.01),缬沙坦组则明显升高(P<0.01)。结论糜酶途径在血管平滑肌细胞AngⅡ生成及细胞增殖中起重要作用。

【Abstract】 Purpose To investigate the influence on the cell proliferation and AngⅡ generation by bloc- king chymase path. Methods The human umbilicus artery smooth muscle cells were cultured and divided into five groups:① DMEM group(blank control group); ② AngⅠgroup;③ AngⅠ plus captopril group; ④ AngⅠ plus chymostatin group; ⑤ AngⅠ plus valsatan group.They are designed to block angiotensin-converting enzyme,chymase and AngⅠrespectively.Each group was undergone cell counting, cell proliferation index and AngⅡ concentration in the culture medium tests. Results The cells’numbers were decreased in the drug interfered groups compared with the Ang Ⅰ group (P<0.01); the cell proliferation of chymostatin and valsatan group were obviously inhibited(P<0.01);the concentration of AngⅡ in chymostatin group was reduced significantly(P<0.01), while it was raised in valsatan group(P<0.01). Conclusions Chymase plays an important role on generation of AngⅡ and proliferation of cultured human vascular smooth muscle cells.
