用流式细胞术评价一种新型人工肝材料的生物相容性

【摘要】 目的通过用流式细胞术(FCM)观察丙稀酰胺接枝改性聚丙烯膜(PP-g-AAm)的生物相容性,来评价FCM在检测医用生物材料的生物相容性中的作用。方法:用材料浸提液培养L929细胞24h后用FCM检测细胞增殖周期;用改性前、后的膜材料分别与新提取的PRP(富含血小板血浆)和PBMC(末梢血单个核细胞)孵育培养后,用FCM分别检测血小板和PBMC的激活标志物CD62P、CD63和CD69。结果:PP-g-AAm组的PI与阴性组及空白对照组比较差异无统计学意义(P=0.063,P=0.053),而与阳性对照组比较差异有统计学意义(P=0.002)。PP-g-AAm膜组CD62P、CD63及CD69的表达率明显少于对照组(P=0.042,P=0.004,P=0.013)。结论:PP-g-AAm无细胞毒性并具有良好的生物相容性,FCM在生物材料的生物相容性评价中有着广泛的应用价值。

【Abstract】 Objective:By using Flow Cytometry (FCM) to detect the biocompatibility of a new bioartificial reactor membrane material (propylene-acidamide grafted polypropylene membrane, PP-g-AAm) in vitro, to assess the value of FCM in detecting the biocompatibility of medical biomaterials. Methods FCM was used to study the proliferative cycle of L929 cells which were cultured with the extraction medium of PP-g-AAm and positive, negative and normal control culture medium respectively after 24 hours. FCM was used to study CD62P and CD63 of the activated platelets after contacting with the PP-g-AAm membranes and ungrafted polypropylene membranes (PP) in PRP (platelet rich plasm) after 30 min. FCM was used to study CD69 of the activated PBMCs (peripheral blood mononuclear cells) after cultured on the PP-g-AAm membranes and PP membranes after 24 hours. Results Although no obvious difference of PI (proliferative index) could be found among the PP-g-AAm group, normal control group and negative group(P=0.063, P=0.053), the PI of the PP-g-AAm group and positive group were significantly different(P=0.002). The ratio of CD62P of the two groups was 5.25±1.42 and 7.63±2.15 respectively, CD63 was 6.01±1.94 and 8.93±2.50 respectively, both were markedly different (P=0.042, P=0.004). The ratio of CD69 of the two groups was 17.24±3.45 and 12.07±2.44 respectively, and differed obviously (P=0.013). Conclusions:The PP-g-AAm membrane has better biocompatibility and no cytotoxicity. And FCM could be used extensively to assess the biocompatibility of biomaterials.
