汉滩病毒核酸疫苗滴鼻及肌注免疫小鼠效果的比较研究

【摘要】 观察汉滩病毒DNA疫苗滴鼻及肌注免疫诱导机体产生的免疫应答,探索汉滩病毒DNA疫苗新的免疫途径。以pcDNA3.1B-S1.3进行肌肉注射及滴鼻免疫小鼠,采用ELISA、淋巴细胞转化试验及特异性CTL杀伤活性测定等方法检测其诱导的系统和黏膜免疫反应的差异。实验结果表明,滴鼻组粪IgA滴度明显高于肌注组(P<0.05);而肌注组血IgG滴度均值虽高于滴鼻组,但两者统计上无显著意义(P>0.05);滴鼻组及肌注组小鼠脾细胞分别经ConA刺激后,其刺激指数作统计分析,结果无显著性差别(P>0.05);肌注组与滴鼻组效应细胞对靶细胞的杀伤力用方差分析进行比较,无显著性差异(P>0.05)。这些均表明,滴鼻免疫对特异的黏膜免疫激发作用明显优于肌肉注射,疫苗的滴鼻免疫途径较肌注途径有着明显的优势。

【Abstract】 To investigate the immune responses induced in BALB/c mice with the DNA vaccine of Hantaan virus (HTNV) by intranasal and intramuscular routes in order to search for the new ways of inoculation with this kind of vaccine, mice were immunized with plasmid pcDNA3.1B-S 1..3 via intranasal or intramuscular routes. The mucosal and systemic immune responses were assayed by ELISA, lymphocyte transformation test and tests for the activities of specific cytotoxic T lymphocyte (CTL). Experimental results showed that the fecal titer of IgA induced via intranasal route was rather higher than that induced via intramuscular route, whereas the titer of IgG in sera by intramuscular route was higher than that by intranasal route, however, there was no significant difference in the latter case. Also, there were no significant differences in the stimulation indices of lymphocytes of spleen cells induced by ConA and in the specific activities of CTL. It appears that the way of immunization by intranasal route might be somewhat better than that by intramuscular route.
