中国新疆维吾尔族、云南彝族KIR多态性分析

【摘要】 分析中国新疆维吾尔族、云南彝族健康人群的KIR基因多态性及单倍型特点。采用序列特异性引物PCR方法(PCR-sequencespecificprimer,PCR-SSP)对维吾尔族、彝族人群基因多态性进行低分辨率检测和单倍型分析。维吾尔族中共检出15种基因型,以AH(5,2)型最常见,频率为22.7%,其次为AF(1,2)和M(2,8),频率分别为13.7%,9.09%。在彝族中共检出19种基因型,以AJ(2,2)型最常见,频率为33.3%,其次为AF(1,2),频率为17.5%。此外,在维吾尔族、彝族中共发现8种新基因型,提示中国少数民族有着自己独特的KIR基因分布频率、单倍型频率和基因型频率。

【Abstract】 To detect the diversity of KIR gene content and the combination of haplotypes in Xinjiang Uygur,Yunnan Yi populations in China, DNA samples from randomly unrelated healthy individuals in Xinjiang Uygur, Yunnan Yi minority are genotyped by SSP/PCR method. All results were compared with each other, and significance of values were assessed by χ2 analysis. Sixteen KIR genotypes have been detected in Uygur population, among which AH (5, 2) was the most common genotype, with the frequency of 22.7%, the later were AF (1, 2) and M(2, 8), with the frequency of 13.7%, 9.09%, respectively. Among Yi population, 21 genotypes have been detected, among which AJ (2, 2) was the most common genotype, with the frequency of 33.3%, the next was AF (1, 2), with the frequency of 17.5%. Furthermore, 8 new genotypes have been found among Xinjiang Uygur, Yunnan Yi minority populations, seven of these genotypes could not be analyzed with the existing 23 heplotypes. These findings suggest that there are distinctive frequencies of KIR gene content and haplotypes, as well as the genotypes in Chinese Xinjiang Uygur, Yunnan Yi minorities.
