小鹅瘟病毒GPV-YG株主要开放性阅读框架核苷酸序列分析

【摘要】 通过PCR技术分段扩增小鹅瘟病毒(Gooseparvovirus,GPV)扬州分离株GPV YG的2个主要开放性阅读框架(openreadingframe,ORF)基因,对扩增产物进行核苷酸序列测定,其结果在GenBank登录。将上述序列与登录的国外参考株GPV-B株作比较,GPV-YG主要ORF核苷酸序列与GPV-B的同源性为94%,显示二者亲缘关系密切。在此基础上,分别确定了GPV-YG编码结构蛋白和非结构蛋白的基因组序列,并推导出GPV-YG结构蛋白和非结构蛋白的相应的氨基酸序列,GPV-YG与GPV B结构蛋白的氨基酸序列同源性为96.4%,非结构蛋白同源性为97.9%,由此进一步分析了GPV-YG结构蛋白和非结构蛋白的氨基酸序列与功能特征。同时,将GPV-YG主要ORF与番鸭细小病毒(MuscovyDuckParvovirus,MDPV)FM作比较,核苷酸序列同源性为80%,非结构蛋白推导氨基酸序列同源性为89.6%,结构蛋白为84.6%。

【Abstract】 The two major open reading frames (ORF) of Goose parvovirus GPV-YG strain were amplified by overlapping PCR. The nucleotide sequence of PCR products was determined. The accession number of this sequence in GenBank is AF416726. The two major ORF’s nucleotide sequence of GPV-YG has a 94% similarity to that of referential strain GPV-B, indicating they are closely related. And the characteristics of GPV-YG structural (VP) and non-structural (NS) proteins were analyzed. The VP proteins of GPV-YG and GPV-B share 96.4% similarity in amino acids sequences, and the NS proteins share 97.9% similarity. The two major ORFs of GPV-YG and MDPV-FM were also compared. They show 80% similarity in nucleotide sequences. The VP proteins of GPV-YG and MDPV-FM share 84.6% similarity in amino acids sequences, and the NS proteins share 89.6%.
