牙本质基质蛋白1、骨桥蛋白及骨涎蛋白在小鼠牙骨质形成中的组织学定位
Histological location of DMP1,OPN and BSP in cementum formation of mouse
【摘要】 目的:研究牙本质基质蛋白1(dentinmatrixprotein1,DMP1)、骨桥蛋白(osteopontin,OPN)及骨涎蛋白(bonesialoprotein,BSP)在小鼠成牙骨质细胞分化及牙骨质形成中的时空分布特点。方法:取出生后第5、10、15、25天的BALB/c小鼠36只,引颈处死,解剖分离含下颌第一磨牙区的下颌骨,新鲜配制4%多聚甲醛固定过夜,甲酸-甲酸钠复合脱钙液脱钙1~3周,常规石蜡包埋,近远中向5μm连续切片,免疫组化PV二步法检测DMP1、OPN及BSP的表达。采用CMIAS2000多功能真彩色病理图像分析系统,高倍镜下每张切片阳性部位沿牙骨质表面随机选取不相重叠的10个视野,计算机测量其积分光密度值,取其均值,计为该指标的1个标本的积分光密度值。相同指标重复3次,均值作为该指标在该标本中的表达强度,各指标在不同发育期的积分光密度值采用重复测量数据方差分析,分别进行统计学处理。结果:DMP1、OPN及BSP在小鼠成牙骨质细胞分化及牙骨质形成过程中呈先后不同的时相表达。DMP1表达最早,出生后第5天在牙囊细胞中表达阳性,一旦牙囊开始分化为牙周膜细胞、成牙骨质细胞和成骨细胞,其表达开始转为阴性;而OPN至出生后第10天出现表达,第15天达到高峰,此后持续表达至牙发育完成的第25天;BSP直到第15天开始表达,持续到第20天后开始减弱,至牙根发育第25天近阴性表达。对3种调节因子的染色积分光密度值的方差分析结果显示:除OPN在第15~25天之间无显著性差异外,其他各因子两两比较差异均具有显著性。结论:牙骨质是由不同序贯发生的多种调节信号顺序启动牙囊细胞分化成牙骨质细胞而形成,DMP1、OPN及BSP在这一过程中先后起着重要的调节作用。
【Abstract】 s] PURPOSE: To study the distribution of dentin matrix protein 1,osteopontin and bone sialoprotein during cementum formation and cementoblasts differentiation in mouse. METHODS: Thirty six BALB/c postnatal mice were divided into four groups according to the developing stages,nine in each was subdivided into three parts randomly to examine the location of DMP1,OPN and BSP,a tooth developing study model was built and examined using histological methods correspondingly. PV two steps immunohistochemical assay was used to localize the distribution of the three regulatory factors timely and spatially. Repeated measures ANOVA and statistical analysis software of CMIAS were used to analyze the intensity of the dyed images. RESULTS: There was a series expression of the three regulatory proteins. DMP1 was expressed in the dental follicle cells in day 5 of postnatal stages and negatively expressed when the cell began to differentiate; meanwhile,since day 10 and on OPN was positively located in cementoblasts until the root developed completely; but BSP was expressed in the same cell in day 15 to 20,and then decreased to be negative. There were significant differences statistically among the three factors in each stage observed in the study except OPN between day 15 and 25. CONCLUSIONS: There are different locations timely and spatially among the three proteins,it hints that they play important different roles during the differentiation of cementoblasts and formation of cementum.
