软骨细胞在几丁糖和透明质酸钠聚合凝胶中培养的实验研究
Experiment of Chondrocyte Culture in Sodium Hyaluronate Augmented Chitosan Hydrogel
【摘要】 目的 探讨几丁糖和透明质酸钠聚合凝胶体作为组织工程软骨细胞支架的可行性。方法 从一月龄兔关节表面削取软骨 ,经酶消化获得软骨细胞。按 0 .8× 10 6 / m L 细胞浓度分别接种于几丁糖凝胶 ,透明质酸钠凝胶 ,几丁糖和透明质酸钠聚合凝胶。通过倒置显微镜观察细胞形态 ,描绘生长曲线 ,甲苯胺蓝染色 ,免疫组化 ,了解软骨细胞生长情况。结果 几丁糖和透明质酸钠聚合凝胶能促进软骨细胞生长、增殖 ,维持软骨细胞表型表达 ,分泌 型胶原及葡萄糖胺上优于透明质酸钠及单纯几丁糖。结论 几丁糖和透明质酸钠聚合凝胶能很好维持软骨细胞正常形态、功能和增殖能力。它可能是组织工程中一种良好的软骨细胞载体材料
【Abstract】 Objective To study the possibility of sodium hyaluronate-augmented chitosan hydrogel(SH-augmented chitosan hydrogel)being used as a scaffold of chondrocytes in tissue engineering.Methods Chondrocytes were isolated by enzymic digestion from the articular cartilage of the one-month rabbits and were cultured in monolayer system.The chondrocytes were respectively seeded in chitosan hydrogel,in sodium hyaluronate hydrogel,in SH-augmented chitosan hydrogel in concentration of 0.8×10 6/mL in vitro.The chondrocytes were observed under light microscope,characteristics of chondrocytes growth were observed with growth curve,toluidine blues staining,type Ⅱ collagen immunohistochemistry reaction.Results All data showed significant improvement in the SH-augmented chitosan hydrogel as compared to the others.Conclusion SH-augmented chitosan hydrogel is good for growth function and phcotype maintenance of chordrocytes.SH-augmented chitosan hydrogel is possible to be used as a good scaffold of chondrocytes in tissue engineering.
