ad-hBMP-2基因转染促进兔下颌骨骨折愈合的实验研究
Bone morphogenetic proteins-2 gene therapy for mandibular fracture union in rabbits
【摘要】 目的研究腺病毒介导的人重组骨形成蛋白-2(ad-hBMP-2)对兔下颌骨骨折愈合的影响。方法16只新西兰白兔随机分成实验组和对照组各8只,建立一侧下颌骨骨折模型后3d,在两组实验动物骨折处注射100μl的ad-hBMP-2病毒液和生理盐水,注射术后4周切取标本,进行X线观察和组织学检查。结果实验组动物骨折处新骨形成量及骨小梁密度显著优于对照组(P<0.05)。结论ad-hBMP-2可明显促进兔下颌骨骨折愈合。
【Abstract】 Objective:To study the effect of bone morphogenetic proteins-2 (ad-hBMP-2) gene therapy on mandibular fracture union.Methods: An animal model of mandibular fracture was established in 16 rabbits which were divided randomly into experimental and control group.100μl ad-hBMP-2 and normal saline were injected respectively into the breaking end of fracture 3 days after operation.All animals were killed 4 weeks after operation and the samples were examined by X-ray and histology.Results: The rate of the new bone formation and the density of the rebirth bone trabecula of the experimental group were superior to the control.Conclusion:ad-hBMP-2 gene therapy can accelerate bone formation during mandibular fracture in rabbits.
