【摘要】 目的研究前列腺素E2(PGE2)对人胃癌细胞株BGC-823低氧诱导因子-1α(HIF-1α)、血管内皮生长因子(VEGF)表达的调节作用。方法以不同剂量的PGE2刺激BGC-823细胞24h,应用RT-PCR和Westerblot方法检测PGE2能否在常氧下诱导BGC-823细胞HIF-1α、VEGF表达。结果PGE2呈剂量依赖性诱导HIF-1α蛋白表达,但对HIF-1αmRNA表达无影响,PGE2呈剂量依赖性诱导细胞VEGFmRNA和蛋白表达。结论PGE2对VEGF表达的诱导可能通过HIF-1α途径介导,这可能是环氧合酶-2(COX-2)诱导肿瘤血管生成的机制之一。更多还原

【Abstract】 Objective:To study the effects of extrinsic PGE₂ on the expression of HIF-1α and VEGF mRNA and protein in human gastric cancer cell line BGC-823 cells.Methods:BGC-823 cells were stimulated by different concentration of PGE₂ for 24 hours.RT-PCR and Western blot were used to investigate the influence of PGE₂ on the expression of HIF-1α and VEGF in the cell line.Results:PGE₂ induced up-regulation of HIF-1α protein and VEGF mRNA and protein in the cell line in a dose-dependent manner,but had no effect on HIF-1α mRNA levels.Conclusion:The effect of PGE₂ simulating the expression of VEGF mRNA and protein may be through HIF-1α pathway in BGC-823 cells,and it may be one of the angiogenesis mechanisms of COX-2.更多还原
