药物治疗的新靶点——细胞凋亡

【摘要】 细胞凋亡是一种由基因调控的细胞程序性死亡过程。多种蛋白质和信号分子参与细胞凋亡过程并进行精确地调控。细胞凋亡可分为caspase依赖性和非caspase依赖性两大途径。其中caspase依赖性途径又分为内源性(线粒体介导)和外源性(死亡受体介导)的凋亡途径。本文对其中一些关键性调控蛋白功能形成初步认识,并尝试把这些蛋白质作为药物治疗的新靶点,借此实现对细胞凋亡过程的干预,为肿瘤、神经退行性疾病、自身免疫性疾病治疗提供新的思路和策略。

【Abstract】 Apoptosis is a programmed cell death that is regulated by genes.A number of proteins and signal molecules are related to the process of apoptosis.These proteins affect each other to regulate the apoptotic process precisely.Inprinciple, there are two main apoptotic pathways:caspase-dependent and caspase-independent pathways.Moreover,the former can be further divided into two alternative pathways that initiate apoptosis:one is mediated by death receptors on the cell surface-sometimes referred to as the“extrinsic pathway”,the other is mediated by mitochondria-referred to as the“intrinsic pathway”. We have rudiment knowledge about roles and functions of some key proteins that control the apoptotic pathway and try to take these proteins as therapy targets to interfere in apoptosis.Therefore,we can develop new ideas and strategies for treatment of several diseases such as tumor,neurodegeneration and autoimmunity.
