【摘要】 目的:建立大鼠绿脓杆菌性角膜炎模型,探讨二硫代氨基甲酸吡咯烷(pyrrolidinedithiocarba-mate,PDTC)对绿脓杆菌脂多糖(lipopolysaccharide,LPS)诱导产生的大鼠角膜炎模型的抗炎作用。方法:建立角膜炎动物模型,模型制作前30min于大鼠球结膜下分别注射生理盐水(炎症组)和PDTC(治疗组),模型制作后1、3、6、12h观察大鼠表现,角膜的组织学改变;免疫组化法检测核因子-κB(nuclearfac-tor-κB,NF-κB)的活化情况。结果:LPS刺激后大鼠角膜组织中NF-κB早期活化。治疗组大鼠角膜中的NF-κBp65核染色阳性细胞明显少于炎症组;鼠眼表现和角膜组织学改变较炎症组改善显著。结论:成功建立了绿脓杆菌性角膜炎动物模型。NF-κB活化在绿脓杆菌性角膜炎的发病中起重要作用,PDTC可有效减轻角膜炎症。更多还原

【Abstract】 Objective:To establish the keratitis model induced by pseudomonas aeruginosa lipopolysaccharide(LPS) in Wistar rats,and to investigate the anti-inflammation effect of pyrrolidine dithiocarbamate (PDTC)on it. Methods:At 30 minutes before LPS exposure, PDTC and normal saline were separatively injected into subconjunctival tissues. At 1,3,6 and 12 hours after LPS exposure, each rat’s clinical representation was observed with slit-lamp microscope, the corneal histopathological structrues were examinded with HE-stained, and the activation and distribution of nuclear factor-κB(NF-κB) were evaluated with immunohistochemical analysis. Results:NF-κB activity in corneas was increased markedly at LPS-induced keratitis in rats.Compared with keratitis group, the positive cells for NF-κB nuclear staining in corneas were significant reduced and the inflammatory symptoms,the histological damages of corneas in PDTC-treatment group were significantly improved. Conclusion:The rat model of pyocyanic keratitis is successfully established. NF-κB activation may play an important role during the pathogenesis of pseudomonas aeruginosa induced keratitis in Wistar rats and PDTC is able to deduce the corneal inflammation.更多还原
