地鼠颊囊黏膜癌变过程中Fas的表达及意义
The expression and significance of Fas in Hamster buccal carcinogenesis
【摘要】 目的:研究Fas蛋白在口腔黏膜癌变过程中的表达及变化规律。方法:用DMBA诱导的金黄地鼠颊囊癌变模型,采用免疫组化SP法检测在黏膜上皮癌变过程(正常黏膜、单纯增生、轻度增生、中度增生、重度增生与鳞癌)中Fas蛋白的表达。结果:Fas在正常地鼠口腔黏膜中广泛表达,在黏膜癌变过程的不同阶段中Fas表达下调,尤其在鳞癌组织中明显降低(P<0.05),且表达部位由细胞胞膜转向胞浆内。结论:Fas参与口腔黏膜肿瘤癌变过程,对口腔癌的早期诊断及治疗提供研究方向。
【Abstract】 Objective: To study the expression of Fas and its change rule in oral carcinogenesis.Methods:Using immunohistochemical method we observed the expression of Fas in DMBA induced Hamster buccal carcinoma including normal control(14),hyperkeratosis(7),epithelial dysplasia(mild 12,moderate 7,severe 9),oral squamous cell carcinoma(OSCC)(25).The expression of Fas was evaluated semi;quantitatively as follows:no expression;mild,<5%positive cells;moderate, 6%~25%positive cells; intense,26%~50% positive cells;very intense,>51%positive cells. Results:Fas was expressed in normal mucosa consistently except stratum basale.However,there was down;regulated in oral dysplasia and OSCC(P<0.05). Furthermore,in OSCC the localization of Fas expression transfered from cell membrane to cytoplasm.Conclusion:The results indicate that the expression of Fas is involved in oral carcinogenesis and that may be related to the cancer cells avoiding the host immune assault, which may be magnificent in oral carcinogenesis mechanism.In the future we will make use of Fas to monitor the degree of epithelial dysplasia and find its potent to malignant transform.Furthermore we can make measures to change the expression of Fas to kill cancer cells,which will be a new and effective method to treat oral cancer.
