EGF对人舌鳞癌细胞株MMP-3表达的影响

【摘要】 目的:探讨EGF对人舌鳞癌细胞株MMP-3表达的影响及其机制。方法:对体外培养的人舌鳞癌细胞株用重组人表皮生长因子(rhEGF)作为刺激因子,采用半定量RT-PCR和WesternBlot方法测定癌细胞中MMP-3mRNA和蛋白含量变化。结果:与未处理组相比,用不同浓度(0.1,1,10ng/ml)EGF处理舌鳞癌细胞后MMP-3表达随浓度的增加而增加,酪氨酸激酶抑制剂Genistein(10mg/ml)能够抑制EGF对癌细胞MMP-3表达的诱导作用。结论:EGF通过酪氨酸激酶通路诱导口腔癌细胞MMP-3的表达。

【Abstract】 Objective: To study the effect of matrix metalloproteinase;3 and its mechanism in tongue squamous cell carcinoma cell line by epidermal growth factor(EGF). Methods: The effects of rhEGF on the production of MMP;3 in tongue squamous cell carcinoma cell were studied by semiquantitative RT;PCR and Western Blot assay. Results: The exposure of tongue squamous cell carcinoma cells to EGF at the indicated concentrations(0.1 ng/ml, 1 ng/ml, 10 ng/ml) increased secretion of MMP;3 in a concentration dependent manner compared with the unstimulated control. We have also found the tyrosine kinase inhibitor Genistein (10 mg/ml)inhibited the increased production of MMP;3 resulting from stimulation with EGF(10ng/ml). Conclusion: EGF is partly involved in the production of MMP;3 via tyrosine kinase signal pathway.
