葡萄籽原花青素降低动脉粥样硬化兔内皮炎症因子水平的研究

【摘要】 目的:探讨葡萄籽原花青素(GSPE)对动脉粥样硬化兔内皮炎症因子水平的影响。方法:30只雄性新西兰大白兔随机分为正常对照(A)、高脂模型(B)、GSPE预防干预(C)组,分别饲喂标准、标准+1%胆固醇、标准+1%胆固醇+1%GSPE的颗粒饲料。于实验开始前1d和实验后第1、2、4、8、12周末取空腹血,用ELISA法检测血清白细胞介素6(IL-6),用EIA法检测血清C反应蛋白(CRP)水平。结果:①IL-6水平:与B组比较,C组IL-6吸光度在8周时明显降低(P<0.05),12周时差异更加明显(P<0.0001);②CRP水平:与B组比较,C组CRP含量在l周末明显降低(P<0.0001)并维持至实验结束;③效应与时间的关系:B组IL-6与时间呈高度相关性(r=0.70314,P<0.0001),C组IL-6与时间无相关关系(P=0.2513)。B组、C组CRP与时间均呈高度相关性(r=0.99047,P<0.0001;r=0.74777,P<0.0001)。结论:GSPE具有降低动脉粥样硬化兔内皮炎症因子水平的作用。

【Abstract】 Objective: To observe the effect of grape seed proanthocyanidin extract (GSPE) on the endothelial inflammation molecule level in atherosclerotic rabbits. Methods: 30 male New Zealand rabbits were randomized equally into three groups, and were given restricted amounts of food (100~120 g/head per day) and free drinking. The control group (group A) was fed on standard diet. The cholsterol group (group B) was fed on standard diet plus 1% cholesterol. The GSPE group (group C) was fed on standard diet plus l% GSPE plus 1% cholesterol. All the rabbits were fed for twelve weeks, and were drawn fastingblood samples on one day before the experiment, at the first, second, fourth, eighth and twelfth weekend of the experiment, then they were determined serum interleukin 6 (IL-6) by commercially available ELISA kits, and serum C-reactive protein (CRP) by commercially available EIA kits. Results: ①Compared with the group B, serum IL-6 absorbency level was lowered markedly by dietary GSPE at the eighth weekend (P<0.05) and even more markedly at the twelfth weekend in group C(P<0.00 01). the serum CRP contents were lowered markedly by dietary GSPE (P<0.00 01) at the first weekend, which lasted to the end of the experiment. ③IL-6 was highly correlated with the treat and time(r=0.703 14,P<0.00 01) in the group B,but not in the group C (P=0.251 3). CRP, in both the group B (r=0.990 47, P<0.00 01) and C (r=0.747 77, P<0.00 01) was highy correlated with the treat and time. Conclusions: GSPE can decrease the endothelial inflammation molecule level in atherosclerotic rabbits, which maybe one of the molecule mechanisms of antiatherogenesis.
