溶栓新药瑞替普酶包涵体纯化和变性条件的研究
Studies on inclusion body purification and denaturation of Reteplase
【摘要】 在溶栓新药瑞替普酶制备的过程中,包涵体的纯化和溶解变性是关键步骤之一.本文探索了包涵体连续三次洗涤纯化方法,得到了较好的纯化效果.比较了两种不同的还原剂(β巯基乙醇和DTT)对包涵体溶解变性的影响,电泳和复性液活性测定结果表明β巯基乙醇是较理想的还原剂.
【Abstract】 The purification and resolution-denaturation of inclusion bodies is a key step in the preparation of recombinant thrombolytic regent Reteplase. Efficient purification is reached by the studied methods. Furthermore, two different reductants, β-mercaptoethanol and DTT, are adapted to resolve and denature inclusion bodies. The results of SAS-PAGE analysis and thrombolytic activity assay of the following refolded Reteplase proteins show that β-mercaptoethanol is more efficient for denaturation than DTT, which indicates that β-mercaptoethanol is an ideal reductant.
