多元有机载体固定化酶水解氨基酸研究
Study on Hydrolysis of Amino Acid with Poly-organic Carrier Immobilized Enzyme
【摘要】 选用反应性单体丙烯酰胺(AM)、N-羟基琥珀酰亚胺(NHM),以N,N’-亚甲基(双丙烯酰胺)(GM)为交联剂,以甲酰胺为制孔剂条件下进行反相悬浮聚合,得到珠状共聚物载体,该载体是一种新的载体形式,在国内外的相关文献中均未见报道.将载体分别用吸附和包埋两种方法固定化中性蛋白酶,测得吸附和包埋两种载体固定化酶量分别为0.328 0和0.026 4 mg/g,固定化酶的活性分别于175.63~484.72μ/g和50.95~82.25μ/g之间变化,比较吸附法和游离态酶水解蚯蚓体蛋白的氨基酸含量,将中性蛋白酶用吸附法固定在所合成的载体上可得到高活性的生物催化剂.
【Abstract】 Taking reactive monomer acrylamide(AM),N-hydroxysuccinimide(NHM) and N,N’-methylene-bis(acrylamide)(GM) as reactants,a copolymer carrier was synthesized with formamide as porogenic agent by inverse suspension copolymerization.This carrier has not been reported until now in the literature.By using the carrier to immobilize neutral albumen enzyme through absorption and embedding,0.328 0 and 0.026 4 mg/g immobilized enzymes were measured respectively,and the activity of immobilized enzymes changed between 175.63~484.72 μ/g and 50.95~82.25 μ/g respectively.Compared with adsorption and hydrolysis of earthworm body protein,the immobilized enzyme has high activity and can be used as an effective biologic catalyst.
