壳聚糖-聚乙二醇接枝共聚物的合成与表征

【摘要】 用1,1’羰基二咪唑活化MPEG法制备活化MPEG,再用活化MPEG与壳聚糖上的伯氨基反应,两步法合成了壳聚糖-聚乙二醇接枝共聚物。用FT-IR、1H-NMR对共聚物的结构进行了表征,分别用重量法、间接法及1H-NMR测定了接枝率约为11%,符合经典隐形纳米粒材料的PEG含量。用X-射线衍射及DSC证明了聚合物的结晶度有所增强,有望作为隐形纳米粒的载体材料使用。为长循环及实体瘤给药系统研究提供新的载体奠定了基础。

【Abstract】 MPEG was modified with 1, 1’-carbonyldiimidazole, then the activated MPEG reacted with primary amino groups of chitosan. Synthesize the graft copolymer of chitosan and polyethylene glycol in two steps. The structure of the copolymer was characterized by FT-IR and~1H-NMR. The graft rate was determined to be 11% with gravimetry method, indirect method and~1H-NMR. It agrees with the PEG content of classical stealth nanoparticles materials. The X-ray diffraction and DSC analyses proved that the crystallinity of the copolymer increased.It is a promising material for the stealth nanoparticles.It is a potential new carrier for the drug delivery systems of long-circulation and solid carcinoma.
