【摘要】 目的　观察蟾酥胶囊含药血清对人肝癌 BEL-7402 细胞凋亡的诱导作用。方法　采用中药血清药理学方法,选择不同浓度含药血清作为实验组,分别与人肝癌 BEL-7402 细胞共同孵育 24h、48h,流式细胞仪检测细胞凋亡率、细胞周期变化及Apo2.7蛋白的阳性表达率;琼脂糖凝胶电泳测定 DNA-Lad der;均与10%无药血清组做对照。结果　流式细胞仪检测可见,各实验组细胞 G1、S期下降,G2/M期升高,产生凋亡峰(sub G1期),Apo2.7蛋白表达的阳性峰明显增高,细胞凋亡率与作用时间、含药血清浓度有一定的时效和量效关系;孵育48h,可见DNA-Ladder出现。结论　蟾酥胶囊含药血清可以诱导人肝癌细胞BEL-7402凋亡,提示蟾酥胶囊是一种有潜力的治疗肝癌药物。更多还原

【Abstract】 Objective To observe the effect on Chansu Capsule with medicated serum in inducing the apoptosis of human hepatocellular carcinoma cell BEL-7402. Methods Using serum pharmacologic method of the traditional Chinese medicine,we chose different concentrations of serum containing chansu capsule as groups,which were respectively co-cultivated with human hepatocellular carcinoma cell BEL-7402 for 24 or 48 hours.The percentage of cell apoptosis,the cycle change of DNA and the level of Apo2.7 protein were examined by flow cytometry; DNA ladder was examined by agar gel electrophoresis;blank serum was taken as control group. Results In study groups,G₁ 、S of cell cycle decreased,G₂/M of cell cycle increased,the level of Apo2.7 protein increased,and the sub-G₁ peaks were observed by flow cytomeric assays;the percentage of cell apoptosis increased with the enhancing of time and concentration,with obvious time -response and dose -response relationship;DNA Ladder appeared by co-culturing 48 hours.Conclusion The chansu capsule could induce apoptosis of human hepatocellular carcinoma cell BEL-7402 and probably become a potential anti-tumor drug.更多还原
