神经生长因子在人胚胎神经管发育的定位表达

【摘要】 目的 研究神经生长因子(NGF)在人胚胎神经管发育过程中的定位表达。方法 用胚胎龄为30天的人胚胎行免疫细胞化学ABC法染色。结果 在人胚胎神经管室带,神经元的细胞浆和细胞核NGF免疫反应阳性;在中间带一部分神经元的胞核NGF免疫反应阳性,另外一部分神经元的胞核 NGF免疫反应阴性,神经元的突起NGF免疫反应阳性;在边缘带NGF的表达与中间带相似。神经管的吻侧 NGF蛋白表达较强,神经管的尾侧NGF蛋白表达较弱。中胚层体节之间可见NGF免疫反应阳性的细胞。结论 NGF是神经管诱导分化的重要信号分子,它通过神经管中间带的NGF水平信号和神经管与中胚层相互作用的垂直信号诱导神经管的发育分化,在神经管的发育中具有十分重要的作用。

【Abstract】 Objective We investigate the expression and the distribution of the neuron growth factors in the development of neural tube of human embryo. Methods The age of human embryo in 30 days and immunocytochemistry ABC technique with anti-NGF antibody were used. Result There were NGF immuno-positive substance in the cytoplasm and nuclei of neuroepithlium in the ventricular zone of neuron tube;There were NGF immuno-positive substance in the nuclei and processes of neurons and some of neurons showed immunoreactive negative in the intennediate zone of neuron tube;The expression of NGF in the margin zone of neuron tube was similar to the intermediate zone of neuron tube.The heavy dense signal of NGF protein was found in the rostrum side of neuron tube and it show relativity weak in the caudal side of neuron tube.The NGF immuno-positive cells were also observed between the somites of embryo under the neuron tube. Conclusion These results might suggest that NGF was important signal molecule to induce neuron tube differentiation,which depend on the plane signal in the intermediate zone of neuron tube and the vertical signal between the neuron and mesoderm,NGF play an important role in promoting and maintaining the biological function of neurons in the development neural tube.
